Abstract:

为构建pGEX-TAT-GFP原核表达质粒并优化GST-TAT-GFP表达条件,将PCR扩增的基因TAT-GFP克隆至质粒pGEX-2T,转化大肠杆菌BL21,IPTG诱导表达并优化表达条件,表达产物进行SDS-PAGE、Western blot及荧光学特性鉴定.结果表明:构建的质粒经PCR、酶切和DNA测序正确,含有重组质粒的宿主菌经过IPTG诱导表达分子量约为54.3 kD的融合蛋白GST-TAT-GFP,并经优化确定最佳的诱导表达条件.
