Abstract:
目的研究连接TAT蛋白转导域的p53融合蛋白对人肝癌细胞株HepG 2的促凋亡作用。方法用反转录法从人正常肝细胞株L-02中获得野生型p53基因cDNA,构建原核表达质粒pET-27b/p53及pET-27b/p53-TAT。IPTG诱导表达重组蛋白,重组蛋白经N-i NTA柱纯化后,加入HepG 2细胞培养液上清,采用四唑氮盐比色法(MTT法)和单细胞凝胶电泳法检测p53及p53-TAT融合蛋白对HepG 2细胞生长的影响。结果成功构建质粒pET-27b/p53及pET-27b/p53-TAT,表达纯化了p53蛋白及p53-TAT蛋白。MTT法和单细胞凝胶电泳法结果表明,与p53蛋白相比,p...
