通过分子克隆与遗传重组技术克隆及表达黑曲霉F0215来源的全部β-半乳糖苷酶,构建了重组菌GS115(p PIC-bgl A)、GS115(p PIC-bgl B)、GS115(pPIC-bglC)和GS115(pPIC-bglE);通过摇瓶发酵和甲醇诱导培养,制备相应的重组β-半乳糖苷酶BglA、BglB、BglC和BglE。重组酶BglA、BglB、BglC和BglE的最适作用温度分别是60,50,60,50℃;最适反应pH分别为4.0,4.5,3.5,4.5;Mg~(2+)、Ca~(2+)、Na~+和Mn~(2+)对上述酶的活性均有促进作用,Cu~(2+)、Fe~(2+)、Ni~(2+)...
