Abstract:
A sensitive method based on capillary electrophoresis ( CE) with amperometric detection (AD) has been developed for the separation and determination of perphenazine and fluphenazine. The working electrode used in here was a 300 mu m diameter carbon disk electrode (vs. Ag/AgCl) operated in a wall-jet configuration. The optimum conditions were obtained with a working potential of 1050 mV and a 75 cm capillary (360 mm, 25 mu m i. d.) at 18 kV in the electrophoretic medium of 20 mmol/L phosphate buffer solution ( pH = 2. 7). Under the optimum conditions, the current response was linear over about two orders of magnitude with detection limits (SIN = 3) of 0. 060 mg/L for perphenazine and 0. 075 mg/L for fluphenazine, respectively. The proposed method has been successfully applied to determine the content of perphenazine and fluphenazine in perphenazine tablet, fluphenazine tablet, spiked urine and rabbit serum samples with satisfactory results.
