Abstract:

本发明提供一种呋喃西林偶联抗原的制备方法及其偶联抗原与应用,其中抗原制备方法包括以下步骤:将呋喃西林溶解于甲醇中,再加入甲醛溶液,呋喃西林与甲醛的摩尔比为1:1~2.5,制成溶液A;将载体蛋白溶解在pH为6.0~7.5的缓冲溶液中,制成溶液B;将溶液A加入到溶液B中,混合均匀后在4~43℃下反应,反应时间为0.5~3d,收集反应液,分离纯化后,得到呋喃西林偶联抗原,其中,呋喃西林与载体蛋白的摩尔比20-120 : 1。本发明所制得的呋喃西林偶联抗原的偶联比可达到15以上,以此呋喃西林偶联抗原免疫小鼠,可产生抗呋喃西林的特异性抗体,此抗体可用于快速检测呋喃西林的含量。
