Abstract:

通过检测小鼠肝脏细胞色素氧化酶 CYP3A11的表达和活性变化,从分子水平探讨银杏叶制剂对肝内药物代谢的影响.利用 RT-PCR 检测银杏叶制剂对小鼠肝脏 CYP3A11以及 PXR 表达水平的影响;采用反相高效液相法测定法测定银杏叶不同组分对小鼠体内硝苯地平代谢速率的影响.银杏制剂明显诱导小鼠肝脏 CYP3A11的表达与活性上升, 银杏黄酮组明显抑制小鼠对硝苯地平的降解速率.银杏黄酮抑制 CYP3A11酶的活性,降低硝苯地平的代谢速率,但银杏叶混合组分激活 CYP3A11的表达,其中总体加速了硝苯地平的代谢.
