Abstract:
目的:研究抗冻多肽对乳酸菌在冷冻干燥过程中的保护作用并探究其保护机理。方法:应用平板菌落计数法测定乳酸菌冷冻干燥前后的活菌数并计算其存活率,利用扫描电镜与红外光谱法分析抗冻多肽对乳酸菌的抗胁迫保护机理。结果:抗冻多肽能够降低乳酸菌冻干损伤,通过对比乳酸菌冻干存活率得到抗冻多肽最佳添加质量浓度为1mg/mL,最适作用pH为8.5,此时添加抗冻多肽的样品组乳酸菌存活率是空白组的4倍。扫描电镜结果显示添加抗冻多肽保护的菌体细胞饱满、完整,空白组则出现皱缩和塌陷。红外光谱结果显示抗冻多肽对脂质体膜有保护作用,可以抑制水化及磷脂的相变和脂质膜的融合。结论:抗冻多肽能与乳酸菌细胞膜磷脂相互作用,降低乳酸菌...
