以研究黑暗链霉菌Tt-49中生物合成基因aprF-G生理功能为目标,利用接合转移法,将同源重组质粒pFD10导入黑暗链霉菌Tt-49,最终获得同源双交换菌株Tt-49G53,并对其生理特性进行考察。结果显示:其生长速度明显比亲株慢,产抗水平比亲株低100倍,仅为13U/mL。采用薄层层析法对其代谢产物进行分析,未检测到安普霉素。
