Abstract:
The exchange capacity and desorption profiles of sisomicin in some macroporous carboxylic acid type ion exchange resins and the conventional sulfonic acid type ion exchange resin were compared. Primary experiments were conducted in three kinds of carboxylic acid type ion exchange resins (Resin I, II, III), and Resin I was selected for further study, and compared the specific characteristics with the sulfonic acid type resin 732. Results showed that the static exchange capacity of Resin- I to sisomicin was 18. 1×107 μg/ml, which was 80% higher than that of sulfonic acid type resin 732. The optimum pH for adsorption of sisomicin was around 7.5; Dynamic exchange capacity of Resin- I was further increased by 13.8% than static exchange capacity. In dynamic exchange of extraction and desorption of sisomicin, the total recovery rate was 82.6%, and it was 8% higher than using the sulfonic acid type resin 732. The elution of sisomicin was performed by use of 5% aqueous ammonia. The elution profile showed that the curve peak of Resin I was more concentrate and less tailing than the sulfonic acid type resin 732.
