A novel sponge-like composite biosorbent fabricated by sodium alginate and polyethyleneimine for uranium(VI) extraction from seawater Scopus

Abstract :

Uranium extraction from seawater (UES) has important strategic significance for maintaining the sustainable development of nuclear energy. This article presents the preparation of a low-cost, efficient, and highly reusable biosorbent sodium alginate/polyethyleneimine (SA/PEI) through a simple one-step crosslinking process. The chemical crosslinking between PEI and SA provides biosorbent excellent mechanical strength and thermal stability. SA/PEI was characterized by using FTIR, XRD, TGA, EDS, XPS, SEM before and after adsorption of uranium. Thermodynamic research results show that the uranium adsorption of SA/PEI is a spontaneous, entropy increasing endothermic process. The adsorption fitted the pseudo-second-order kinetic model and Langmuir model with maximum adsorption capacity reach 353.09 mg g−1, illustrating that the adsorption mechanism is monolayer chemical adsorption. The interaction between SA/PEI and uranium is synergistic chelation by amino and carboxyl, which is consistent with the results calculated by DFT. After 14 days of adsorption in 100 L natural seawater, the adsorption capacity of SA/PEI was 3.58 mg g−1, with an average adsorption efficiency of 0.256 mg g−1 day−1, which is faster than most reported alginate adsorbents. The cost of using SA/PEI to UES is $168 per kilogram of uranium. These results indicate that SA/PEI hydrogel has great potential in practical seawater application. © 2024 Elsevier B.V.

Abstract :

In this work, we synthesized a novel beta-cyclodextrin/poly(2-acrylamide-2-methylpropionic acid)/attapulgite (CAMA) hydrogel for uranium(VI) adsorption. Various instrumental techniques such as FTIR, SEM, EDX, and TGA were utilized for characterization of the CAMA. Simultaneously, different important parameters, in specific pH, time, initial uranyl ion concentration and temperature were investigated to evaluate the optimal conditions of the adsorption process. The experiments proofs that the adsorption follows the pseudo-second-order kinetic model so as the Langmuir model, and revealed the maximum adsorption capacity of 434.78 mg g(-1) at 298.15 K. Regeneration studies further sighted out the good reusability of the synthesized CAMA hydrogel.

Abstract :

Extracting uranium from seawater is of great significance for ensuring the sustainable development of nuclear energy. This article designs a simple and mass-produced porous sponge polyamidoxime/polyethyleneimine (PAO/PEI) for uranium extraction from seawater. The prepared porous sponge has excellent flexibility and mechanical strength, which can withstand harsh marine environments. In a 50 mg/L uranyl solution, the maximum adsorption capacity of PAO/PEI is 666.67 mg/g. Moreover, PAO/PEI sponge exhibits excellent selectivity for uranium in competitive adsorption conditions. FTIR and XPS are used to explore the adsorption mechanism, and the results showed that the synergistic effect of amidoxime and amino groups enhanced the binding of uranium(VI). The adsorption capacity after 15 days of enrichment in 100L natural seawater is 9.79 mg/g, and the average rate is 0.653 mg/g/day. The results indicate that PAO/PEI porous sponge has broad application prospects in large-scale seawater uranium(VI) extraction.

Abstract :

Sensitive quantification of protein biomarkers is highly desired for clinical diagnosis and treatment. Yet, unlike DNA/RNA which can be greatly amplified by PCR/RT-PCR, the amplification and detection of trace amount of proteins remain a great challenge. Here, we combined allosteric probe (AP) with magnetic bead (MB) for assembling an on-bead DNA synthesis system (named as APMB) to amplify protein signals. The AP is designed and conjugated onto the MB, enabling the protein biomarker to be separated and enriched. Once recognizing the biomarker, the AP alters its conformation to initiate DNA synthesis on beads for primary signal amplification. During the DNA synthesis, biotin-dATPs are incorporated into the newly synthesized DNA strands. Then, the biotin-labeled DNA specifically captures streptavidin (STR)-conjugated horseradish peroxidase (HRP), which is used to catalyze a colorimetric reaction for secondary signal amplification. By using carcinoembryonic antigen (CEA) as a protein model, the APMB can quantify protein biomarkers of as low as 0.01 ng/mL. The response values measured by APMB are linearly related to the protein concentrations in the range 0.05 to 20 ng/mL. Clinical examination demonstrated good practicability of the APMB in quantifying serum protein biomarker. The on-bead DNA synthesis could be exploited to improve protein signal amplification, thus facilitating protein biomarker detection of low abundance for early diagnosis.

Abstract :

Rapid and sensitive quantification of pathogenic bacteria is highly desired for environmental health supervision and food safety control. Yet, the amplification and detection of bacteria with a concentration lower than 10(2) cfu mL(-1) remains a great challenge. Here, we combined an allosteric aptamer (AAP) with a gold nanoparticle (AuNP) for assembling a bridge-DNA synthesis system (named as AuNP-BDS) to amplify the bacterial signals. The AAP and its paired primer (PP) were covalently linked to two different AuNPs, respectively: one named as AAP-AuNP and the other PP-AuNP. Upon recognition of the antigen from the pathogenic bacteria, AAP alters its conformation to initiate DNA synthesis on the AuNP surface. The DNA products from AAP-AuNP and PP-AuNP form bridges to each other through base pairing, resulting in the aggregation and colorimetric response of the AuNPs. By using E. coli O157:H7 as an example, the AuNP-BDS could quantify pathogenic bacteria in water with a concentration as low as 10 cfu mL(-1) within 60 min and without any enrichment. The colorimetric response values of AuNP-BDS were found to be linearly related to the bacterial concentrations in the range of 10 to 10(3) cfu mL(-1). Good practicability of the AuNP-BDS in quantifying E. coli O157:H7 from tap water, juices, and milks was demonstrated. The AuNP-BDS could be exploited to facilitate the rapid and sensitive quantification of pathogenic bacteria for food safety control.

Abstract :

The separation and enrichment of uranium from various resources such as seawater is of great significance to the sustainable development of nuclear energy. Therefore, there is an urgent need to develop an adsorbent with low cost and easy availability, simple preparation process, and environment friendly. In this work, we used inexpensive natural polymer material (sodium alginate) and natural adsorbents (beta-cyclodextrin and diatomite) to prepare a novel biosorbent beta-cyclodextrin/sodium alginate/diatomite (CSD) hydrogel beads for uranium adsorption by a simple method. The results of FTIR, XRD, SEM, EDX and XPS characterization proved that the CSD were prepared. Static adsorption experiments showed that pH, contact time, initial concentration of U(VI) and temperature has significant impact on the adsorption capacity of CSD. Adsorption fitted the pseudo-second-order kinetic model and Langmuir model and the adsorption mechanism is chemical adsorption. In the presence of other metal ions (Cu2+, Mg2+, Ca2+, Na+, K+), CSD has obvious selectivity for uranium(VI) adsorption. In addition, the elution and reusability studies show that CSD has excellent reusability. Overall, CSD is an inexpensive, green preparation, efficient, and environment friendly biosorbent with obvious selectivity for U(VI). It has potential application in U(VI) enrichment from seawater or salt lake water.

Abstract :

There are currently many materials for treating residual antibiotics in the environment, but none of them can specifically remove antibiotics. In addition, these materials are not sensitive enough to low concentrations of antibiotics. Here we show that a sensitive and specific material was developed by the preparation of magnetic Fe3O4-PAMAM-antibody complexes for treating tetracycline. The prepared antibody complexes can specifically treat tetracycline from aqueous solutions and the tetracycline removal ability by adsorption was also investigated. Controlled experiments were carried out with the effects of solution pH, temperature, and initial concentration of the tetracycline. The tetracycline was completely removed within 35 min at room temperature 30 degrees C with the maximum removal rate of almost 100%. Therefore, this material for specifically combining antigen and antibody to treat tetracycline indicated good application prospects for the waste water treatment.

Abstract :

The emerging and development of green chemistry has once again drawn the researchers' attention to eliminating the use and generation of hazardous materials. Here we report the use of a safe and effective fixative, chlorine dioxide (ClO2), instead of traditional hazardous fixatives for the cross-linking of cellular proteins to improve immunofluorescence staining of bacteria. The concentration of ClO2 needed for 100% fixation is 50 mu g ml(-1), which is much lower than that of traditional fixatives (1000-10000 mu g ml(-1)). The ClO2 mediated cross-linking can preserve the integrity of bacterial cells and prevent cell loss through lysis. Meanwhile, lysozyme can permeabilize the bacterial cells, allowing the labelled antibodies to diffuse to their intracellular target molecules. By using E. coli O157:H7/RP4 as a gram-negative bacteria model, immunofluorescence staining assays for both intracellular protein and surface polysaccharide were carried out to investigate the effect of ClO2 fixation on the staining. The results demonstrated that ClO2 fixation could prevent the target antigens from cracking off the bacteria without damage on the interaction between the antibodies and antigens (either for polysaccharide or protein). As a safe and effective fixative, ClO2 has potential practical applications in immunofluorescence staining and fluorescence in situ hybridization for single bacteria/cell analysis.

Abstract :

Enzyme-linked immunosorbent assay (ELISA) is an economic and easy operation technique that has been widely used for the detection of protein in industry. However, the low loading capacity of the enzyme reporter has contributed to the low sensitivity of traditional ELISA, and the cross-linking procedures of the enzyme-labeled antibody in ELISA methods can lead to the inactivation of the enzyme, which will further decrease the sensitivity. To address this issue, herein we fabricated "carrier-free" nanoparticles to obtain a horseradish peroxidase (HRP) labelled reporter with a high HRP loading capacity. A disulphide-containing bis-N-hydroxy succinimide (NHS) crosslinker (NHS-SS-NHS) was used to control the link and release of traceless HRPs, thus without reduction of its enzymatic activity. The HRP nanoparticle (NanoHRP) was successfully applied for dot blotting and ELISA. When carcinoembryonic antigen (CEA) was used as a target, the detection limit of the NanoHRP-based ELISA was 0.005 ng mL(-1), which was about 400 times more sensitive than traditional ELISA. A good correlation between the CEA concentrations and the response values measured by NanoHRP ELISA was obtained in the range of 0.005 to 1 ng mL(-1). This concept could be exploited to improve ELISA tests, especially those requiring a high accuracy, to facilitate physicians in deciding the appropriate medical treatment.

Abstract :

Uranium in the environment can damage human health and ecosystem. There is a need for excellent adsorbents to remove U(VI) from aqueous solutions. Here we synthesized a novel beta-cyclodextrin/poly(acrylic acid)/permutite (CAP) hydrogel composite by a simple method. Physicochemical characterizations of the materials were conducted by XRD, FTIR, SEM, EDX, and TGA. The effect of pH value, contact time, initial U(VI) concentration, and temperature were researched. A pseudo-second-order kinetic model, intra-particle diffusion model, and Langmuir isotherm model were used to describe the U(VI) adsorption behavior, and the maximum adsorption capacity of U(VI) was 833.33 mg/g at 25 C. Thermodynamic analysis showed that the adsorption process of U(VI) was endothermic and spontaneous. Furthermore, the excellent reusability indicated that CAP hydrogel composite could be potentially used as a promising sorbent for the removal of U (VI) in wastewater.
