Abstract :

The processes of ferroptosis and drug-induced liver injury (DILI) correlate closely to oxidative stress in mitochondria which will affect intracellular microenvironmental parameters such as lysosomal viscosity. Therefore, it is of great significance to dynamically monitor ONOO- in mitochondria and viscosity in lysosomes for understanding redox signal transduction but remains challenging. Here we report a near-infrared fluorescent probe, HCy-OH-mito, which has the dual-organelle targeting ability for the detection of ONOO- in mitochondria and viscosity in lysosomes via a "three-key-and-lock" strategy. HCy-OH-mito has been successfully applied to independently and simultaneously label mitochondria and lysosomes in live cells, and monitor the fluctuations of ONOO- in mitochondria and viscosity in lysosomes during ferroptosis and APAP-induced liver injury and in live zebrafish.

Abstract :

Mercury (Hg) is an exceedingly toxic heavy metal which leads to grievous nonbiodegradable contamination and human health threat. Thus, developing a practical, quick, effective and labor-saving method for detecting Hg2+ is of great significance. In this study, we rationally design two novel near-infrared (NIR) monothiosquaraine-based probes MTSQ-1 and MTSQ-2 for tracking and bioimaging of Hg2+ with the mercury-desulfurization strategy. MTSQ-1 and MTSQ-2 show ultrahigh sensitivity, superb selectivity and rapid response for spotting of Hg2+ in vitro. We find that it is the intramolecular hydrogen bond between the carbonyl oxygen of the squaraine core and the ortho-hydroxyl substituent of the anilinyl moiety that promotes the desulfurization of the probes, inducing enhanced fluorescence emission. Related sensing mechanism has been fully discussed through experimental methods and theoretical calculations. Meanwhile, MTSQ-1 has been used in detection of Hg2+ in river samples, and MTSQ-2 encapsulated in beta-CD possesses good ability for imaging of Hg2+ in HeLa cells with high signal-tobackground ratio.

Abstract :

Adenosine 5'-triphosphate (ATP), as a "molecular currency" for intracellular energy transfer, not only participates in various physiological activities, but also involves in various pathological processes of living cells. Therefore, the establishment of a simple, rapid, sensitive and accurate ATP analysis method is of great significance for the study of its physiological functions. In this work, we rationally design a near-infrared phenylboronic acid group-functionalized dicyanovinyl squaraine (SQ- PBA3), with which a supramolecular assembly was constructed for fluorescent imaging of ATP. In PBS buffer, SQ-PBA3 changes self-assembly when adding cetyltrimethyl ammonium bromide (CTAB) deriving fluorescence response of SQ-PBA3 to ATP with the maximum emission at 700 nm, due to the specificity of phenylboronic acid to diols and the multiple electrostatic interactions between SQ PBA3, ATP and CTAB molecules. Meanwhile, the probe was successfully applied to monitor intracellular ATP level in MCF-7 cells with high sensitivity and selectivity.

Abstract :

This article describes the rational design and synthesis of a monothiosquaraine (MTSQ) fluorescent Hg2+ probe that exhibits near-infrared fluorescence, ultrahigh brightness, fast response, high sensitivity and excellent selectivity. Utilizing the strategy of the incorporation of the heavy atom effect and self-assembly caused quenching, the background fluorescence of MTSQ was greatly lowered and the sensitivity in PBS buffer solution was highly enhanced with a very low limit of detection (LOD = 1.2 nM). Systematic studies revealed excellent selectivity and specificity for Hg2+ over other thiophilic metal ions such as Ag+, Pb2+, and Cd2+. The mechanistic investigation indicates that self-assembly and desulfurization of MTSQ are responsible for Hg2+-triggered significantly higher fluorescence enhancement. With high sensitivity, optimal pH range of 3-7, weak background, good water-solubility and low cytotoxicity, MTSQ therefore can be applied for imaging Hg2+ in live cells and zebrafish. The very simple but effective strategy reported here can provide a more convenient approach for designing NIR probes.

本发明公开了一种苯硼酸修饰的近红外方酸染料及其制备方法和应用,其是将吲哚修饰的苯硼酸衍生物和二氰基乙烯基方酸衍生物混合,经溶解、回流、减压蒸除溶剂及硅胶柱层析纯化,制得所述近红外方酸染料。该方酸染料具有较好的稳定性及优异的光学性能,尤其是对位取代的苯硼酸基团,可与ATP分子中的顺式二醇部分结合,故可通过染料、ATP和CTAB分子之间的多重静电作用,引发染料吸收光谱和荧光光谱的改变,从而可作为ATP检测的荧光探针,用于细胞内ATP的荧光成像检测,且该探针具有较好的生物相容性和细胞通透性、较低的生物毒性,对ATP具有较好的检测灵敏度。

Abstract :

Proteins are among the pivotal constituents in living systems. The development of fluorescent probes with appropriate selectivity and sensitivity at biologically relevant signaling levels offers opportunities to explore their roles in physiological functions. Here we report the design and synthesis of a new squaraine based fluorescent molecule SQ1 and the study of its sensing interaction with bovine serum albumin (BSA). The interaction of SQ1 with BSA was investigated by fluorescence and UV-vis absorption spectroscopy. Displacement studies were carried out with site selective probes to locate the binding site, showing that the SQ1 binds at site I. The results obtained from the docking studies are also in good agreement with the experimental results. The SQ1cBSA was used for cell incubation and in vitro fluorescence imaging studies. 本发明公开了一种水溶性近红外单硫代方酸染料及其制备方法和应用,其制备方法包括以下步骤:(1)将氧代方酸染料和劳森试剂混合,溶于适当的溶剂,N2保护下回流数小时;(2)冷却至室温,减压除去溶剂,得粗产品;(3)经硅胶柱层析纯化,得近红外单硫代方酸染料产物。该单硫代方酸染料稳定性好,光学性能优异,位于中心四元环的硫原子可以有效地捕获溶液中的汞离子,引发染料吸收光谱和荧光光谱的改变,故可作为检测汞离子的荧光探针。氧醚链侧链的引入有效地提高了探针在纯水体系的溶解度,并且具有较好的生物相容性和细胞通透性,较低的生物毒性,可用于细胞及斑马鱼活体内汞离子的成像检测,且表现出较好的汞离子检测灵敏度。

本发明公开了方酸染料及其在比色荧光双响应铜离子探针制备的应用,其制备方法包括以下步骤:(1)将苯胺衍生物和方酸混合,溶于适当的溶剂,接上分水器,N2保护下减压回流分水数小时;(2)将步骤(1)所得的反应混合物冷却至室温,减压除去溶剂,得粗产品;(3)粗产品经硅胶柱层析纯化,得近红外方酸染料产物。该方酸染料稳定性好,光学性能优异,侧链上的氮原子和氧原子与铜离子有较好的结合力,在体系中加入各种金属离子中,只有铜离子导致染料的荧光猝灭且溶液颜色由蓝色变成无色,因此该染料可作为铜离子检测的比色/荧光探针,可用于含水介质中的铜离子检测,表现出较好的检测灵敏度。

Abstract :

In this work, a novel fluorescent chemosensor, based on a polyether bridged squaraine, has been developed for the detection of Pb2+, and gives excellent sensitivity and selectivity using a cation-induced disaggregation strategy. This probe exhibited a specific "turn on" fluorescent response to Pb2+ with a detection limit of 1.70 x 10(-8) M, and the identification for Pb2+ was not impacted by competing ions such as Na+, Li+, Mg2+, Ca2+, Co2+, Ag+, Cd2+, and Hg2+. Due to the high binding ability of the polyether bridged ionophore toward Pb2+, the disaggregation process could be trigged by the formation of squaraine-Pb2+ complex, resulting in a fast fluorescent release. The sensing mechanism has been confirmed by NMR, IR, and dynamic light scattering experiments. Further research shows that this novel fluorescent probe can be applied in the fast detection of Pb2+ in urine and shrimp samples. (C) 2016 Elsevier B.V. All rights reserved.

Abstract :

A near-infrared fluorescent chemosensor based on squaraine has been synthesized, and its cascade recognition for copper ion, biological phosphate PPi, and amino acids has also been investigated by spectroscopic techniques. SQ-1 exhibits high selectivity and sensitivity for sensing Cu2+ over other metal ions by reversibly forming a [SQ-1-Cu2+] complex. By tandem ligand exchange, PPi and amino acids can remove Cu2+ from the complex and restore the spectral signal of SQ-1, which can be used for cascade recognition of these biologically active species. Notably, an IMPLICATION logic gate can be constructed based on this bifunctional chemosensing system. (C) 2016 Elsevier B.V. All rights reserved.
