Pluronic F127 nanoparticles modified with folate and loaded with phycocyanobilin for tumor-targeted photodynamic therapy SCIE

Abstract :

Photodynamic therapy (PDT) has emerged as a promising cancer treatment strategy due to its minimally invasive nature and high specificity for malignant tissue. Our previous work has shown that phycocyanobilin (PCB), a natural food coloring derived from Arthrospira Platensis, has the potential to be a photosensitizer. Compared to most reported photosensitizers, PCB is almost non-toxic and readily available. However, its water insolubility and inadequate targeting delivery to cancer cells have been identified as significant obstacles, which compromise its bioavailability and further clinical application. In the present study, Folate-modified pluronic F127 nanoparticle drug delivery system (FF) was developed, which possessed specific tumor targeting, homogeneity, dispersibility and good biocompatibility. This system has been shown to enhance PCB uptake by 4T1 mouse breast carcinoma cells, increase the intracellular ROS production, induce 4T1 cell apoptosis in a concentration-dependent manner, and significantly improve the anticancer activity of PCB. The IC50 value of Phycocyanobilin-Folate acid-pluronic F127 nanoparticle drug delivery system (PFF) under light irradiation was determined to be 42.75 +/- 0.78 mu mol/L, which was 26.62 % higher than that of PCB alone. Zn-Phthalocyanine is a typical second-generation photosensitizer. The therapeutic index (TI) value of PFF was found to be 8.09 under light irradiation, which was significantly higher than that of Zn-Phthalocyanine (TI = 1.08). In vivo studies have shown the efficacy of PFF in delivering PCB to tumor sites, resulting in a significant inhibition rate of tumor growth inhibition (96.15 +/- 3.70 %). Furthermore, PFF has been demonstrated to exhibit excellent biocompatibility, suggesting its potential for use in clinical treatments.

Abstract :

Rubropunctatin is a natural and harmless secondary metabolite of the fungus Monascus with promising anticancer properties. However, its water insolubility affects its distribution and absorption in vivo, its bioavailability and further application in the clinic is limited. Herein, a core-shell nano-micelle Rubropunctatin Deoxycholate Sodium/Poloxamer 188/Phospholipid nano-micelle (RDPP-MNPs) was prepared by the film evaporation. RDPP-MNPs exhibited uniform size (152.73 f 1.55 nm), low CMC (0.027 f 0.010 mg/mL) and pHsensitive sustained release, which effectively prolonged the circulation time of the drugs in vivo. The results of CCK-8 cytotoxicity experiment showed that RDPP-MNPs could significantly inhibit the activity of Hela and 4T1 cells (IC50 = 7.73 f 0.09 mu g/mL, IC50 = 9.25 f 0.19 mu g/mL), respectively. In addition, RDPP-MNPs have a greater ability to induce apoptosis of Hela cells by promoting the uptake of drugs into cells in a concentrationdependent manner. Finally, we verified the tumor inhibitory effect and biosafety of RDPP-MNPs by tumorbearing mouse model. The tumor inhibition rate was up to 71.49 f 3.53 % with no systemic toxicity, validated by organ histology. These results showed that the therapeutic efficacy of Rubropunctatin could be significantly enhanced by DPP-MNPs. And based on its good anti-cancer activity, bioavailability and biosafety, it has potential in gentle chemotherapy and can reference other natural compounds in cancer therapy.

Abstract :

The inadequate vascularization and abnormal immune microenvironment in the diabetic bone defect region present a significant challenge to osteogenic regulation. Inspired by the distinctive characteristics of healing staged in diabetic bone defects and the structure-function relationship in the natural periosteum, we fabricated an electrospun bilayer biomimetic periosteum (Bilayer@E) to promote regeneration of diabetic bone defects. Here, the inner layer of biomimetic periosteum was fabricated using coaxial electrospinning fibers, with a shell incorporating zinc oxide nanoparticles (ZnO NPs) and a core containing silicon dioxide nanoparticles (SiO2 NPs) mimicking the cambium of periosteum; the outer layer consisted of randomly aligned electrospun fibers loaded with deferoxamine (DFO), simulating the fibrous layer of periosteum; finally, epigallocatechin-3-gallate (EGCG) was coated onto the bilayer membrane to obtain Bilayer@E. The presence of EGCG on the Bilayer@E surface efficiently triggers a phenotypic transition in macrophages, shifting them from an M1 proinflammatory state to an M2 anti-inflammatory state. Moreover, the sequential release of ZnO NPs, DFO, and SiO2 NPs exhibits antimicrobial characteristics while coordinating angiogenesis and promoting osteogenic mineralization in cells. Importantly, the biomimetic periosteum shows strong in vivo bone tissue and periosteal regeneration properties in diabetic rats. The integration of sequential drug release and immunomodulation, tailored to meet the specific healing requirements during bone regeneration, offers new insights for advancing the application of biomaterials in this field.

Abstract :

In anterior cruciate ligament (ACL) repair methods, the continuous enzymatic erosion of synovial fluid can impede healing and potentially lead to repair failure, as well as exacerbate articular cartilage wear, resulting in joint degeneration. Inspired by the blood clot during medial collateral ligament healing, we developed a composite scaffold comprising collagen (1 %, w/v) and polyvinyl alcohol (5 %, w/v) combined with platelet-rich plasma (PRP). The composite scaffold provides a protective barrier against synovial erosion for the ruptured ACL, while simultaneously facilitating tissue repair, thereby enhancing the efficacy of ACL repair techniques. The composite scaffold is primarily formed through hydrogen bonding between molecular chains and physical cross- linking of microcrystalline regions using a simple cyclic freeze-thaw method, resulting in improved mechanical properties and an extended degradation period. The maximum tensile fracture load of the composite scaffold reached 5.99 +/- 0.30 N. The incorporation of PRP facilitates cell migration, proliferation, and blood vessel growth by enabling slow release of various growth factors. In vivo results demonstrate that this composite scaffold promotes rabbit hindlimb rupture ACL healing by stimulating fibroblast proliferation, collagen deposition, microvascular formation, and proprioceptor generation. Furthermore, it effectively reduces meniscus and cartilage wear while mitigating bone arthritis and joint degenerative diseases. Overall, our proposed composite scaffold holds great promise as a candidate for ACL healing.

Abstract :

Pressure ulcers, resulting from prolonged external pressure or shear forces on skin and underlying tissues over bony prominences, lead to tissue ischemia and impaired lymphatic drainage. Without timely intervention, these wounds can progress to severe complications including cellulitis, chronic infections, and osteomyelitis. In this study, we developed a chitosan/sodium (3-glycerophosphate/gelatin (CS/(3-GP/GEL) thermosensitive hydrogel system to enhance the therapeutic efficacy of adipose-derived mesenchymal stem cells (ADSCs) in pressure ulcer healing. The incorporation of gelatin addresses the limitations of conventional CS/(3-GP hydrogels, such as low mechanical strength and poor biocompatibility. The optimized CS/(3-GP/GEL hydrogel exhibits good inject-ability, suitable gel formation time and pH, facilitating efficient ADSCs encapsulation. Furthermore, the hydrogel demonstrates good water absorption capacity, degradability, and rheological properties, making it suitable for biomedical applications. In vitro studies confirmed the hydrogel's high cytocompatibility, supporting ADSCs viability and proliferation. Additionally, the CS/(3-GP/GEL@ADSC composite demonstrated pro-angiogenic properties, suppressed reactive oxygen species-mediated apoptosis, and facilitated the accumulation of M2-type macrophages. In vivo evaluations revealed that the CS/(3-GP/GEL@ADSC composite had high histocompatibility and accelerated pressure ulcer healing in a rat model, mediated through enhanced angiogenesis, M2-dominant macrophage polarization, and improved extracellular matrix remodeling. These findings highlight the potential of CS/(3-GP/GEL@ADSC as a promising therapeutic strategy for pressure ulcer management.

Abstract :

The redox homeostasis defense mechanism of tumor cells is one of the prime reasons for the unsatisfactory effect of photodynamic therapy (PDT). So far, little attention has been paid to this obstacle. In this work, we reported a synthesizing simple yet versatile nanogel (BCPS), synthesized by cystamine dihydrochloride functionalized sodium carboxymethylcellulose (CMC-SS), bovine serum albumin, and Phycocyanobilin self-assembly. The BCPS reduced the levels of glutathione molecules by reacting with glutathione, thereby interfering with intracellular redox homeostasis and enhancing the sensitivity of tumor cells to PDT. The BCPS was shown to possess excellent serum stability, high blood compatibility, low toxic side effects, and higher reactive oxygen species (ROS) utilization. After irradiation, the BCPS could significantly increase intracellular ROS level by approximately 1.6-fold and decrease the IC50 to HeLa cells by approximately 1.5-fold, compared to the pre-functional drugs BCP. This proposed strategy, based on increasing the utilization rate of ROS in tumor cells is promising for application potentials in tumor therapy.

Currently, cisplatin resistance has been recognized as a multistep cascade process for its clinical chemotherapy failure. Hitherto, it remains challenging to develop a feasible and promising strategy to overcome the cascade drug resistance (CDR) issue for achieving fundamentally improved chemotherapeutic efficacy. Herein, a novel self-assembled nanoagent is proposed, which is constructed by Pt(IV) prodrug, cyanine dye (cypate), and gadolinium ion (Gd3+), for systematically conquering the cisplatin resistance by employing near-infrared (NIR) light activated mild-temperature hyperthermia in tumor targets. The proposed nanoagents exhibit high photostability, GSH/H+-responsive dissociation, preferable photothermal conversion, and enhanced cellular uptake performance. In particular, upon 785-nm NIR light irradiation, the generated mild temperature of approximate to 43 degrees C overtly improves the cell membrane permeability and drug uptake, accelerates the disruption of intracellular redox balance, and apparently enhances the formation of Pt-DNA adducts, thereby effectively overcoming the CDR issue and achieves highly improved therapeutic efficacy for cisplatin-resistant tumor ablation. A novel self-assembled nanoagent (Cy-Pt@HA NP) is developed to conquer the cascade drug resistance (CDR) issue of cisplatin. The proposed nanoagent presents NIR light-triggered mild hyperthermia, superior cellular uptake, and tumor-microenvironment-responsive dissociation for effective ablation of drug-resistant tumors, thereby opening a new avenue for addressing CDR issue and maximizing the platinum-based therapeutic efficacy for cancer treatment.image

The management of severe full-thickness skin defect wounds remains a challenge due to their irregular shape, uncontrollable bleeding, high risk of infection, and prolonged healing period. Herein, an all-in-one OD/GM/QCS@Exo hydrogel was prepared with catechol-modified oxidized hyaluronic acid (OD), methylacrylylated gelatin (GM), and quaternized chitosan (QCS) and loaded with adipose mesenchymal stem cell-derived exosomes (Exos). Cross-linking of the hydrogel was achieved using visible light instead of ultraviolet light irradiation, providing injectability and good biocompatibility. Notably, the incorporation of catechol groups and multicross-linked networks in the hydrogels conferred strong adhesion properties and mechanical strength against external forces such as tensile and compressive stress. Furthermore, our hydrogel exhibited antibacterial, anti-inflammatory, and antioxidant properties along with wound-healing promotion effects. Our results demonstrated that the hydrogel-mediated release of Exos significantly promotes cellular proliferation, migration, and angiogenesis, thereby accelerating skin structure reconstruction and functional recovery during the wound-healing process. Overall, the all-in-one OD/GM/QCS@Exo hydrogel provided a promising therapeutic strategy for the treatment of full-thickness skin defect wounds through actively participating in the entire process of wound healing.

Abstract :

Platelet-rich plasma (PRP) has become an important regenerative therapy. However, the preparation method of PRP has not been standardized, and the optimal platelet concentration for PRP used in skin wound repair is unclear, leading to inconsistent clinical efficacy of PRP. Therefore, the development of standardized preparation methods for PRP and the investigation of the dose-response relationship between PRP with different platelet concentrations and tissue regeneration plays an important role in the development and clinical application of PRP technology. This study has developed an integrated blood collection device from blood drawing to centrifugation. Response surface methodology was employed to optimize the preparation conditions, ultimately achieving a platelet recovery rate as high as 95.74% for PRP (with optimal parameters: centrifugation force 1730x g, centrifugation time 10 min, and serum separation gel dosage 1.4 g). Both in vitro and in vivo experimental results indicate that PRP with a (2x) enrichment ratio is the most effective in promoting fibroblast proliferation and skin wound healing, with a cell proliferation rate of over 150% and a wound healing rate of 78% on day 7.
