干制、储存条件及模拟消化对龙眼干中5-羟甲基糠醛含量的影响

Abstract :

为探究龙眼干在加工、储存过程中5-羟甲基糠醛(5-hydroxymethylfurfural,5-HMF)的生成规律及安全性,测定不同温度干制、储存过程中龙眼干5-HMF含量的变化,以及干制过程中还原糖、总酸、总游离氨基酸和褐变度的变化,并分析各种变化之间的相关性;然后通过体外模拟消化实验及肠道上皮细胞模型对其进行安全性分析.结果表明:随着干制温度的上升,5-HMF在干制温度大于85 ℃时显著升高(P<0.05),5-HMF含量与总酸含量和褐变度呈正相关,与总游离氨基酸及还原糖含量呈负相关;龙眼干在冷藏(4 ℃)、常温(25 ℃)、高温(37 ℃)储存条件下,5-HMF随储存时间延长先上升后下降,不同条件下的增长趋势不同,其中25 ℃条件下最快开始下降且总体含量较低;龙眼干中的5-HMF含量在口腔消化阶段短暂升高,在胃、肠消化阶段显著降低;龙眼干水提液能抑制5-HMF的吸收,且对于24 mmol/L 5-HMF的Caco-2细胞毒性有一定的抵消作用.结果说明,优化龙眼干的干制和储存条件能够降低5-HMF的含量;正常食用的情况下,人体消化道能降低龙眼干中5-HMF,提高其食用安全性.

Abstract :

透明质酸(HA)是一种非硫酸化糖胺聚糖,2021年获批为新食品原料后,引起食品科研人员与产业界的广泛兴趣。近年来,越来越多的研究显示,HA的生物活性根据其分子质量的不同,发挥促炎或抗炎、促细胞增殖或抗增殖作用。为了高效、精准地提高HA在食品中的应用价值,有必要根据分子质量的差异性,全面归纳总结HA在食品相关领域的研究进展。本文概述HA的应用开发进展,重点分析不同分子质量HA的制备方法、细胞功能、消化吸收特性,从维持稳态、缓解炎症两个维度分析HA对肠道的调节作用,以期促进HA在营养与健康产业的精准开发及应用。

Abstract :

To explore the formation pattern of 5-hydroxymethylfurfural (5-HMF) and the safety of dried longan during processing and storage, this study monitored the changes in 5-HMF content during drying and storage at different temperatures. The changes in the contents of reducing sugar, total acid and total free amino acid and browning degree during processing were determined and their correlation with 5-HMF content was analyzed. Moreover, the safety of dried longan was analyzed using in vitro simulated digestion experiments and a Caco-2 cell model. The results showed that as the drying temperature increased above 85 ℃, the 5-HMF content significantly increased (P © 2024 Chinese Chamber of Commerce. All rights reserved.

Ameliorative Effect of Orally Administered Nano-Superoxide Dismutase Liposomes on Ulcerative Colitis in Mice EI CSCD PKU

Abstract :

With an increasing incidence rate,ulcerative colitis (UC) is an idiopathic inflammation of the colon causing diarrhea and blood in the stool. Existing studies have shown that oxidative stress is an important factor leading to UC, while superoxide dismutase (SOD) has a certain effect on relieving UC symptoms. To improve its bioavailability, superoxide dismutase (SOD) was self-assembled into nanoparticles (ΔSOD) by heating it. The activity, transmembrane capacity and radical scavenging capacity of ΔSOD were significantly better than those of native SOD. ΔSOD liposomes (L-ΔSOD) were prepared by reverse-phase evaporation and orally administered to mice with UC. The results showed that compared with the positive control and model groups, L-ΔSOD significantly ameliorated UC, reduced disease activity index (DAI) score, restored the morphology of colon tissue, increased the length of colon, reduced the density of colon and spleen coefficient, and restored the integrity of mucosal epithelium. Besides, L-ΔSOD could reduce the contents of malondialdehyde (MDA) and pro-inflammatory factors in colon tissue and increase the levels of glutathione peroxidase (GSH-Px) and tight junction protein mRNA expression. These results confirmed that oral L-ΔSOD has a good ameliorative effect on UC, which will lay the foundation for the application of SOD in promoting human health. © 2023, China Food Publishing Company. Abstract :

Oxidative stress is generally considered inseparable from the development and exacerbation of ulcerative colitis (UC). Therefore, reducing oxidative stress has become a possible way to alleviate UC. In this study, the therapeutic effects of different doses of liposome-embedded superoxide dismutase (L-SOD) on mice with DSS-induced UC were systematically investigated. The results showed that L-SOD significantly attenuated the signs of colitis in mice, including colonic shortening, diarrhoea, bloody stools, and histopathological changes. L-SOD ameliorated DSS-induced oxidative damage, increased SOD, catalase (CAT), and glutathione (GSH) activities, and decreased malondialdehyde (MDA) levels. In addition, L-SOD ameliorated the inflammatory response by inhibiting the expression of myeloperoxidase (MPO) and pro-inflammatory cytokines and protected barrier function by promoting the expression of the tight junction proteins occludin and ZO-1 in the colon. Importantly, the results demonstrated a bell-shaped distribution of therapeutic effects relative to the administered dose, with an optimal dose of 150 000 U kg(-1). These results indicate that L-SOD has great potential as an ingredient in functional foods for the prevention and mitigation of UC.

粗粮速食米替代高脂饮食对2型糖尿病小鼠糖脂代谢的影响 CSCD PKU

粗粮速食米(instant rice with added coarse grain,IR/CG)是一种新型的粗粮制品,既具有自热米饭食用方便的特点,又具有粗粮谷物的营养价值,但其产生的健康影响并不明确,因此本研究通过高脂饮食(high fat diet,HFD)联合链脲佐菌素腹腔注射诱导小鼠产生糖尿病,探究IR/CG对2型糖尿病(type 2 diabetes mellitus,T2DM)小鼠糖脂代谢的作用。通过IR/CG与高脂饲料不同比例混合制备小鼠饲料,低、中、高剂量(25%(质量分数,下同)、50%、75%IR/CG)饲料以及100%IR/CG饲料分别记作IR/CG-L、IR/CG-M、IR/CG-H及IR/CG,利用上述饲料喂养小鼠,并与基础饲料、高脂饲料喂养的小鼠(分别记作Control、Model组)进行生理、生化指标等的比较。在饮食干预期间测定小鼠体质量、空腹血糖浓度、摄食量及葡萄糖耐量;IR/CG饮食干预4周后,计算小鼠脏器系数,对小鼠肝脏、胰腺进行苏木精-伊红染色,测定小鼠空腹胰岛素水平、血糖血脂相关指标,并测定小鼠促炎细胞因子水平。结果显示,IR/CG的摄入能够不同程度改善T2DM小鼠高血糖血脂水平,其中IR/CG-H、IR/CG组改善效果较为显著。此外,IR/CG-H、IR/CG能够显著改善小鼠胰岛素抵抗现象,减少胰岛素分泌、降低胰高血糖素及糖基化血红蛋白水平,减轻小鼠炎症状态。通过对肝脏、胰腺的脏器指数及病理学切片分析发现,IR/CG干预能够有效减轻小鼠肝脂肪变性,部分恢复胰岛结构。上述结果表明IR/CG替代HFD对T2DM小鼠具有显著的调节血糖血脂作用,本研究能为粗粮自热米饭产品及粗粮功能性食品的开发提供理论依据。

溃疡性结肠炎(ulcerative colitis,UC)是一种特发性结肠炎症,会引起腹泻、便血、腹痛等症状,其发病率不断上升.现有研究表明氧化应激是造成UC的重要原因之一,而超氧化物歧化酶(superoxide dismutase,SOD)在缓解UC症状上有一定的效果.因此,本实验通过加热SOD使其自组装形成SOD纳米颗粒(记作ΔSOD),以提高SOD的生物利用度,通过理化指标分析发现ΔSOD纳米颗粒的活力、细胞跨膜能力以及自由基清除能力较天然SOD均显著提高;再采用逆向蒸发法制备ΔSOD脂质体(记作L-ΔSOD),并灌胃UC模型小鼠,结果显示,与阳性对照组和模型组相比,L-ΔSOD可以显著改善UC,降低小鼠疾病活动指数评分,恢复小鼠结肠组织状态,增加结肠长度,降低结肠密度和脾脏系数,恢复黏膜上皮完整性;同时L-ΔSOD可以降低结肠组织MDA含量,并通过减少细胞促炎因子质量浓度、提高谷胱甘肽过氧化物酶活力和紧密连接蛋白mRNA水平来改善UC.综上,L-ΔSOD对UC具有较好的改善作用,本实验可为SOD促进人体健康的实际应用提供参考.

The chitosan-coated alginate gel particles (CS/SA) and chitosan-coated alginate-shellac gel particles (CS/SA/Lac) were constructed to serve as a potential oral delivery system to enhance the stability and activity of superoxide dismutase (SOD) in this study. The encapsulation efficiency (EE), enzyme activity retention ratio (ER), particle structure, and SOD release behavior, as well as the effect of shellac, were evaluated. The best formula is determined by the percentage of total effective activity (PTEA).The results showed that CS/SA prepared by 0.30 parts per thousand chitosan, 1.00% CaCl2, 2.00% sodium alginate (M/G = 1:1, Mw: 38,857 kDa), and gelation solution pH 5.50 had the best chitosan shell, the strongest gel structure and the maximum PTEA which was 10.75 +/- 1.45%. Based on the optimal formula, addition of shellac that formed shellac-Ca2+ microspheres in the gel network was averse to SOD encapsulation, but it significantly improved the stability of SOD in gastric conditions. By adding 15.00% shellac, the PTEA reached 16.30 +/- 1.64%. At optimal formulation, SOD encapsulated in CS/SA was completely released in the stimulated intestinal fluid and accumulative release enzyme activity (EA) was 966.62 +/- 103.75 (U) after 6 h. However, the CS/SA/Lac with a low swelling ratio only released SOD in stimulated colonic fluid and the EA value reached 1692.28 +/- 101.82 (U) after 1 h. These findings demonstrated that CS/SA and CS/SA/Lac were excellent candidates for SOD oral administration, which was useful in the food and biomedical industries for developing functional products containing SOD.

Instant rice with added coarse grain (IR/CG) is a new type of coarse grain product which is self-heating and convenient to eat and has the nutritional value of coarse grains. However, its health effects are not clear yet. Therefore, in this study, the effect of IR/CG on glucose and lipid metabolism was explored in mice with type 2 diabetes mellitus (T2DM) induced by high fat diet (HFD) combined with intraperitoneal injection of streptozotocin. The mice were fed on HFD alone (model), its mixture with IR/CG in different ratios: 25% (low-dose IR/CG, IR/CG-L), 50% (medium-dose IR/CG, IR/CG-M), and 75% (high-dose IR/CG, IR/CG-H), IR/CG alone, or a basal diet (control). During the dietary intervention, the body mass, fasting blood glucose, food intake and glucose tolerance of the mice were measured. After the IR/CG intervention for four weeks, the organ coefficients were calculated, and the pathological changes in liver and pancreas tissues were examined by hematoxylin and eosin (HE) staining. Fasting insulin levels, blood glucose and lipid levels, and proinflammatory cytokines levels were determined. The experimental results showed that dietary supplementation of IR/CG improved blood glucose and lipid levels in T2DM mice to different extents, and this effect was more prominent in the IR/CG-H and IR/CG groups. In addition, IR/CG-H and IR/CG significantly mitigated insulin resistance, decreased insulin secretion, glucagon and HbA1c levels, and reduced oxidative stress injury and inflammatory cytokine levels in T2DM mice. By analyzing the organ index and pathological sections of liver and pancreas tissues, it was found that IR/CG intervention could effectively alleviate liver steatosis and partially restore the structure of the pancreatic islets in mice. Therefore, the substitution of high-fat diet by IR/CG has a significant effect on regulating blood glucose and lipids in T2DM mice, which can provide a theoretical basis for the development of self-heating rice products and functional foods with coarse-grain. © 2023 Chinese Chamber of Commerce. Ulcerative colitis (UC) is an idiopathic intestinal disease with an increasing incidence, and all the therapeutic drugs have different degrees of side effects. Studies have found that collagen peptide can relieve the symptoms of UC, and is safe and healthy. However, the current research is mainly limited to the prevention effect of UC, and there is still a lack of research on the therapeutic effect of UC. Eel skin is rich in collagen, but eel head is generally discarded in processing, resulting in a waste of resources. Meanwhile, there is a lack of research on eel skin collogen hydrolysates. In this study, collagen was extracted from eel skin and hydrolyzed in the hope of obtaining eel skin collagen hydrolysate (CH) with therapeutic effect on UC, so as to improve the utilization value of eel processing by-products. It was found that eel skin contained abundant collagen, accounting for 27.87% of the wet weight of the skin. The extracted collagen was of high purity and mainly type I collagen, and eel skin could be used as a high - quality source of collagen. Collagen was enzymatically hydrolyzed to obtain eel skin collagen hydrolysate (CH) with antioxidant activity, and CH was administered to DSS induced UC mouse model. The results showed that CH could safely and healthily reduce DAI score to 0 and improve colon status in UC Model mice compared with Positive control group and Model group. Pathological sections of colon tissue showed that CH could restore the integrity of mucosal epithelium and reduce inflammatory cell infiltration. Oxidative stress index of colon tissue showed that CH could reduce MDA content (0.75 nmol/mg protein) and MPO activity (0.11 U/g tissue) in colon tissue, and improve the antioxidant capacity of colon tissue and reduce oxidative stress level by increasing GSH content (22.92 umol/g pro) and activities of SOD (693.61 U/g pro), CAT (1.69 U/g pro) and GPx (141.29 U/g pro) antioxidant enzymes, so as to treat UC. © 2023 Chinese Institute of Food Science and Technology.
