Abstract :
In this work, the acute and subchronic toxicities of desaminotyrosine (DAT) by oral administration in SD rats and its effects on the intestinal microflora were investigated. The acute toxicity test showed that DAT is a low-toxic substance with a LD50 of 3129 mg/kg. The subchronic toxicity test showed that DAT has no toxicity at a low dose (125 mg/kg/day). However, DAT exhibited obvious toxicities to food intake, liver, kidney, and lung at higher dose (250 mg/kg/day and 500 mg/kg/day). DAT inhibited the food intake of rats in a dose-dependent manner. Serum biochemical analysis showed that DAT can increase the serum glucose level of rats. Fecal microbiota analysis showed that DAT treatment can significantly change the intestinal microflora of rats, the dose of 125 mg/kg/day has the most significant effect on the diversity of intestinal microbiota. In daily application, the side effects caused by DAT might be gastrointestinal irritation, weight loss, liver or kidney injury, and blood sugar elevation. Based on our study, the no-observed-adverse-effect level (NOAEL) of DAT is 125 mg/kg BW/day for rats.

Abstract :
Traditional methods for aflatoxin B1 (AFB1) detection are complex, time-consuming, labor-intensive, and high cost. Moreover, they require sophisticated large-scale instrumentation, which limits their on-site rapid detection. Herein, phycocyanin fluorescent nanospheres based on fluorescence immunochromatographic assay were developed for quantitative detection of AFB1 at parts-per-billion (ppb) levels in foodstuffs. Phycocyanin and antiAFB1 monoclonal antibodies were coupled on the surface of latex nanospheres to amplify the fluorescence signal and improve the sensitivity. The fluorescence intensity was measured by a self-developed smartphone-based reading system. Under the optimal conditions, this approach achieved quantitative point-of-care detection of AFB1 within 25 min. The calibration curve for AFB1 was linear in the range of 0.2-48 ppb, and the limit of detection was 0.16 ppb. The practical applicability of the proposed approach was demonstrated by the determination of AFB1 in naturally contaminated samples, and the results were consistent with HPLC detection.

Abstract :
Acute full-thickness wounds require amore extended healing period, thus increasing the risk of infection. Severe infection frequently resulted in wound ulceration, necrosis, and even life-threatening complications. Here, a hybrid hydrogel comprising aminated collagen (AC), oxidized sodium alginate (OSA), and antimicrobial peptides (polymyxin B sulfate and bacitracin) was developed to enhance full-thickness wound healing. The AC with low immunogenicity and high biocompatibility was made from marine fish scales, which are eco-friendly, low-cost, and sustainable. The cross-linked hydrogel was formed by a Schiff base reaction without any catalysts and additional procedures. As expected, the presented hybrid hydrogel can effectively against E. coli and S. aureus, as well as promote cell growth and angiogenesis in vitro. In addition, the hydrogel can promote full-thickness wound healing in a rat model through accelerating reepithelialization, collagen deposition, and angiogenesis. Our work demonstrated that the hybrid hydrogel has promising applications in the field of wound healing, which would prompt the utilization of marine fish resources during food processing. Background: It has been demonstrated in some studies that triterpenoid acid extract fromEriobotrya japonica leaf is beneficial to prevent hyperlipidemia or insulin resistance. However, the effect of triterpenoid acids in Eriobotrya japonica leaf on a series of typical symptoms of metabolic syndrome (MetS) has been rarely studied systemati-cally. Therefore, the present study aims to systematically evaluate the effect of Eriobotrya japonica leaf triterpenoid acids (ELTA) on MetS and explore its potential mechanism. Methods: ELTA (HPLC purity 95.2 %) was prepared and administered orally (200 mg/kg) to C57BL/6 J mice fed with a high-fat diet (HFD) for 12 weeks. Pioglitazone (30 mg/kg) was used as a positive control drug. Food intake, body weight, total lipid in feces, lipid profiles, inflammatory factors in serum, hepatic glutathione, and lipid peroxide were measured. Oral glucose tolerance test (OGTT) and insulin tolerance test (ITT) were performed to evaluate insulin sensitivity. RT-qPCR and molecular docking were performed to explore the potential mechanism. Results: ELTA administration reduced body weight gain, relative liver weight, and relative visceral adipose weight. The levels of serum total cholesterol, triglycerides, low-density lipoprotein cholesterol, hepatic total cholesterol, and hepatic triglycerides were also reduced. ELTA reduced the area under curve (AUC) of blood glucose curves in OGTT and ITT. Relative mRNA level analysis of genes related to MetS showed that ELTA can effectively increase the transcriptional levels of Nrf2, HO-1, PPAR-gamma, GluT2, GK, FXR, while effectively decrease those of PTP1B, p65, TNF-alpha, IL-6, SREBP, 11 beta HSD-1. Molecular docking showed that the ligands in ELTA can bind to 11 beta HSD-1, GK, PPAR-gamma, and JNK, the important targets involved in MetS. Conclusions: ELTA can effectively alleviate visceral central obesity, insulin resistance, dyslipidemia, oxidative stress, and inflammation of HFD-induced MetS in C57BL/6 J mice. This is possibly achieved by acting on 11 beta HSD-1, GK, PPAR-gamma, and JNK.

Wound dressings with long-term antimicrobial activity are highly desired for treatment of chronic wound infections. Herein, the sustained antimicrobial wound dressings were developed by using antibiotic agents, ciprofloxacin HCL (CIP) and gentamicin sulfate (GS), covalent bonding to natural polymer matrix composites, carboxymethyl chitosan (CMC) and collagen (COL). By amide bond formation between antibiotic agents and polymer chains, two antimicrobial wound dressings CMC-COL-CIP and CMC-COL-GS were prepared. The presented wound dressings exhibited high water absorption capacity, excellent water vapor transmission rate (WVTR), appropriate mechanical properties, and impressive stability. Cytocompatibility of the dressings was demonstrated by in vitro human skin fibroblast (HSF) cells culture study. The results of in vitro and in vivo studies indicated that the two antimicrobial wound dressings have effective antimicrobial activity and prolonged antimicrobial period. Furthermore, the antimicrobial dressings could promote the wound healing, reepithelialization, collagen deposition, and angiogenesis. It also displays superiority wound healing effects compared to commercially available silver-based dressings (Aguacel Ag). This work indicates that the prepared antimicrobial wound dressings have great potential application in chronic wound healing, such as severe wound cure and diabetic foot ulcers. Herein, an autonomous table-tennis-motion-type DNA machine (ATT DNA machine) was proposed to integrate target recycling amplification (TRA) and triple strand-displacement amplification (T-SDA) into one signaling system for the efficient amplification detection of p53 gene. Firstly, X-shaped DNA duplex probe (XSP) was formed via the simple annealing of Probe A and Probe B. Upon addition of p53, the hybridization of p53 with Probe A results in the peeling of Probe B from XSP. Then, the primer binds to the Probe B to trigger a circular strand-displacement amplification, during which new primers are generated. Moreover, the newly-produced primers can in turn activate the target recycling amplification and triple strand-displacement amplification via bouncing back and forth between different Probe As and Probe Bs in a manner analogous to the table tennis motion. This newly-developed DNA machine can be used to detect p53 down to 0.5 pM with an excellent specificity, offering a 100-fold enhanced sensitivity compared to a non-amplification one-to-one hybridization based system. Moreover, this DNA machine is able to be applied for target analysis in complex environments. Additionally, the potential for practical applications is also verified, demonstrating a promising alternative tool for early diagnosis of cancers.

In the human genome, single-nucleotide polymorphisms (SNPs) can often lead to the activation of oncogenes and have a close association with tumor development or progression. Specific identification of SNPs and quantitative analysis of oncogenes could provide insight into the basis for early diagnosis and risk assessment of malignancy, as well as personalized medicine. Herein, we proposed a new biosensing platform to screen SNPs through the ligation/polymerization-mediated signaling of a split molecular beacon (SMB) where target-recycling benefits can be achieved. The SMB consists of two fragments, one of which contains a fluorophore group and the other was modified with a quencher group. In the absence of target DNA, the two fragments hybridize with each other and the fluorescence is quenched. After exposure to a wild-type target, they can be opened and ligated together by the ligase using the target as the template. Moreover, the primer is able to bind to target hybridization-released binding sites and initiate strand-displacement amplification (SDA), generating an enhanced signal. By combining the specificity of nick ligation with the signal amplification of target recycling, the human K-ras oncogene as a model target can be detected down to 8 pM. Moreover, a wide dynamic range (more than 3 orders of magnitude) and excellent ability to discriminate single-base mutations are achieved. Due to the homogeneous reaction and simple experimental steps, the target assay is greatly cost-efficient and readily automated. The desirable features and advantages make the proposed genotyping strategy a promising platform for genetic analysis and molecular diagnostics.

The purpose of the current study was to construct a -cyclodextrin drug carrier for rubropunctatin to improve its water solubility and light stability for future cytotoxicity studies. The inclusion complexation behavior of rubropunctatin with -cyclodextrin was investigated using FESEM, FT-IR and XRD. A molecular docking study was performed to elucidate the most probable inclusion structure. The inclusion complex could be completely dispersed in water and had a small size of 121.87 +/- 2.13 nm (n = 3), a good PDI (0.320 +/- 0.017), and an acceptable potential value of -27.7 +/- 0.32 mV (n = 3). Furthermore, the stability of the rubropunctatin in water under light irradiation was found to be greatly enhanced after being encapsulated in cyclodextrin, and it exhibited a retention rate of over 70% vs. 10.17%. In addition, the cytotoxicity of the inclusion complex was evaluated by MTT assay and Annexin V-FITC/PI detection using cervical adenocarcinoma HeLa cells. The results showed that the inclusion complex had comparable toxicity compared to rubropunctatin solubilized with 0.4% DMSO. More importantly, the formation of the inclusion complex contributed greatly to the intensification of the bioavailability of rubropunctatin because the use of organic solvent was avoided.

Phycocyanin (PC) is a natural fluorescent protein which is widely used in medical diagnosis, biology, biotechnology, foods, and medicine. However, the disadvantages in traditional purification methods, such as complicated preparation process, low yield, inadequate quantity, and high price, limit its range of application. In this study, we established a rapid purification method suitable for large-scale production to prepare PC from Arthrospira platensis. The membrane separation and two-stage ammonium sulfate precipitation yielded PC with a purity ratio (A(620nm)/A(280nm)) of 3.65. This purification method of PC proved to be efficient and cost-effective compared with previously reported methods. Sodium dodecyl sulfate polyacrylamide gel electrophoresis (SDS-PAGE) analysis showed that the molecular weight of alpha and beta subunits of PC was 16.6 and 21.2 kDa. The main molecular weight of intact PC determined through high-performance size exclusion chromatography (HPSEC) was around 91 kDa, indicating that the PC from A. platensis existed mainly in the form of (alpha beta)(3) at pH 7.0. The purified PC fluorescent probe was compatible for light-emitting diode (LED)-charge-coupled device (CCD) fluorescence density strip qualitative detection system. A relatively sensitive novel fluorescent test method was developed for the quantitative detection of PC showing a linear response to PC within the range of 0.07-3.64 mu M, and with a limit of detection of 0.034 mu M. This method could provide expedient quantitative information and showed great potential for rapid detection in environmental and food safety research.
