Updated toolkits for nucleic acid-based biosensors SCIE

Abstract :

Much progress has been made in the biosensor area with the prosperity and development of nucleic acid nanotechnology and molecular biology. Especially, the advances made in dynamic DNA nanotechnology and the increasing sophistication of toehold-mediated strand displacement reactions have put new energy into nucleic acid-based biosensor designs. The exposure of the gene-editing ability of clustered regularly interspaced short palindromic repeats (CRISPR) associated proteins (Cas) has also greatly revolutionized the way nucleic acid can be operated in biosensing systems, providing versatile strategies for signal handling. Hence, in this review, we will discuss the application of these two technologies as the expanded tools for nucleic acid-based biosensors and focus on their strategies governing signal trans-duction. Besides, technologies like DNA computing and deep learning approaches which flashed in nucleic acid biosensors very recently are also discussed in the perspective and outlook part.(c) 2023 Elsevier B.V. Abstract :

Salmonella is one of four key global causes of diarrhea, and in humans, it is generally contracted through the consumption of contaminated food. It is necessary to develop an accurate, simple, and rapid method to monitor Salmonella in the early phase. Herein, we developed a sequence-specific visualization method based on loop-mediated isothermal amplification (LAMP) for the detection of Salmonella in milk. With restriction endonuclease and nicking endonuclease, amplicons were produced into single-stranded triggers, which further promoted the generation of a G-quadruplex by a DNA machine. The G-quadruplex DNAzyme possesses peroxidase-like activity and catalyzes the color development of 2,2'-azino-di-(3-ethylbenzthiazoline sulfonic acid) (ABTS) as the readouts. The feasibility for real samples analysis was also confirmed with Salmonella spiked milk, and the sensitivity was 800 CFU/mL when observed with the naked eye. Using this method, the detection of Salmonella in milk can be completed within 1.5 h. Without the involvement of any sophisticated instrument, this specific colorimetric method can be a useful tool in resource-limited areas.

Abstract :

Rapid and accurate detection of Salmonella is extremely important to ensure food safety. Combining nucleic acid amplification with CRISPR/Cas12a can realize sensitive and specific detection of foodborne pathogens. However, present CRISPR/Cas-based methods mainly depend on fluorophore and quencher dual-labeled ssDNA as the reporter, which brings in high cost and steric hindrance. Herein, a new signal output format has been developed. The method takes advantage of graphene oxide adsorbing FAM-ssDNA and quenching the fluorescent emission while CRISPR/Cas12a recovering the fluorescent emission by trans-cleaving the FAM-ssDNA. Factors affecting the performance of the sensor have been optimized, and it is shown that the oligonucleotide of 12 nt with the fluorophore modified at the 3 ' end displays the highest signal to noise ratio. The signal output method has been combined with recombinase polymerase amplification for detection of Salmonella and presents consistent specificity and sensitivity (5 x 101 copies) as real-time PCR. By replacing the dual-labeled ssDNA reporter with FAM-ssDNA and GO, this signal output method saves similar to 65% cost. Notably, this method does not require a real-time thermal cycler or any other sophisticated instrument. Besides pathogens, this easy and convenient method can also be extended to detect other targets.

Abstract :

Bacterial infection of wounds is one of great concern to patients, and rapid and correct detection of bacterial infection is crucial to ensure accurate diagnosis and early intervention. Based on the principle that glucose can only be consumed by live bacteria, a fluorescent biosensor was constructed to detect four kinds of common infectious bacteria (Acinetobacter baumannii, Escherichia coli, Pseudomonas aeruginosa and Staphylococcus aureus) in skin wounds, taking advantage of magnetic bead-aptamer for recognizing, sorting and enrichment, platinum nanoparticles for signal amplification. The linear detection range of AB, EC, PA and SA were 27- 8 x 106 CFU/mL, 10- 2.5 x 107 CFU/mL, 34- 2.5 x 107 CFU/mL and 10- 1.0 x 107 CFU/mL, respectively, and the limits of detection were 27 CFU/mL, 10 CFU/mL, 34 CFU/mL and 2 CFU/mL. Furthermore, all target bacteria in samples containing 8 x 108 CFU/mL of other interfering bacteria have been successfully identified and quantified. The proposed method was also successfully applied to the detection of bacterial infection in skin wounds of mouse and human, including the detection of separate bacterial infection as well as a coinfection. The recovery of this method was in the range of 90.161- 109.961%. Thus, this proposed method can be a promising candidate for rapid and convenient evaluation of infectious bacteria in point-of-care settings.

Multiplex detection methods for mycotoxins in agricultural products: A systematic review SCIE

Abstract :

The combined contamination of multiple mycotoxins (MTs) is a matter of utmost concern, posing significant threats to both the economy stability and the well-being of humans and animals. Consequently, there is an urgent demand for highly sensitive and portable multiplex detection methods to effectively identify and quantify MTs. The primary objective of this paper is to provide a comprehensive overview of the methods to detect multiple MTs in agricultural products, including instrumental and biosensor approaches published in the last five years. Specifically, we emphasize detection methods for antibody (ab)- and aptamer (apt)-based biosensors, while undertaking a comparative analysis of their performance, particularly focusing on the sensitivity. Furthermore, this review proposes several promising technologies that can be leveraged in the future and outlines the challenges and prospects associated with achieving rapid, accurate, and intelligent detection of MTs.

Nucleic acid aptamer-based nanomicelles have great potential for nanomedicine and nanotechnology applications. However, amphiphilic aptamer micelles are known to be inherently unstable upon interaction with cell membranes in the physiological environment, thus potentially compromising their specific targeting against cancer cells. This flaw is addressed in the present work which reports a superstable micellar nanodelivery system as an amphiphilic copolymer self-assembled micelle composed of nucleic acid aptamer and polyvalent hydrophobic poly(maleic anhydride-alt-1-octadecene) (C18PMH). Using Ce6 as a drug model, these C18-aptamer micelles exhibit efficient tumor-targeting and -binding ability, facilitating the entry of Ce6 into targeted cells for photodynamic therapy. In addition, they can be loaded with other hydrophobic drugs and still demonstrate favorable therapeutic effects. As such, these C18-aptamer micelles can serve as a universal platform for loading multiple drugs, providing a safer and more effective solution for treating cancer.

Abstract :

Food packaging plays a vital role in the food supply chain by acting as an additional layer to protect against food contamination, but the main function of traditional conventional packaging is only to isolate food from the outside environment, and cannot provide related information about food spoilage. Intelligent packaging can feel, inspect, and record external or internal changes in food products to provide further information about food quality. Importantly, intelligent packaging indicators will account for a significant proportion of the food industry's production, with promising application potential. In this review, we mainly summarize and review the upcoming progress in the classification, preparation, and application of food packaging indicators. Equally, the feasibility of 3D printing in the preparation of intelligent food packaging indicators is also discussed in detail, as well as the limitations and future directions of smart food packaging. Taken together, the information supported in this paper provides new insights into monitoring food spoilage and food quality.

Until now, corona virus disease 2019 (COVID-19) remained to be an enormous threat for global health. As one viral illness induced by severe acute respiratory syndrome coronavirus 2 (SARS-CoV-2), versatile, rapid and sensitive method for SARS-CoV-2 detection in early stage is urgently needed. Here, we reported an ultrasensitive and visual in-one-tube detection method which could be accomplished within half an hour from sampling -to-result. By integrating all reactions in one tube, liquid handling steps were omitted and amplicon contamina-tion could be totally avoided. Magnetic beads were employed to achieve the fast extraction of viral nucleic acid and increase the sensitivity. Using portable thermocycler and blue light, the fluorescent results could be directly observed by naked eyes. The proposed method is of higher specificity and sensitivity, nearly at single molecule

Surimi-based products (such as fish ball, kamaboko and crabstick) are well accepted by consumers and have broad market prospects owing to their health and nutritional properties and convenience. The surimi-based product is an elastic gel formed based on the gelation properties of myofibrillar protein. The processing procedure includes chopping, heating and exogenous substance addition. Therefore, changes in the structural and functional characteristics of surimi myofibrillar protein under typical processing conditions can affect the quality of surimi-based products. In this article, the quality changes and control methods of surimi myofibrillar protein under the typical processing conditions of physical field (heating and comminution), chemical field (pH and ionic strength) and biological field (enzyme and microorganism) are reviewed, aiming to provide a theoretical basis for further development of the surimi-based product processing industry. © 2022, China Food Publishing Company. Abstract :

This study aimed to investigate the protective effects of ganoderic acids (GA) from Ganoderma lucidum against liver injury and intestinal microbial disorder in mice with excessive alcohol intake. Results showed GA supplement significantly inhibited the abnormal elevation of the liver index, serum lipid parameters, aspartate aminotransferase and alanine aminotransferase in mice exposed to alcohol intake, and also significantly protected the excessive lipid accumulation and pathological changes. Alcohol-induced oxidative stress in the liver was significantly ameliorated by GA intervention through reducing the levels of maleic dialdehyde and lactate dehydrogenase and increasing the levels of glutathione, catalase, superoxide dismutase and alcohol dehydrogenase. Intestinal microbiota profiling demonstrated GA intervention modulated the composition of intestinal microflora by increasing the levels of Lactobacillus, Faecalibaculum, Romboutsia, Bifidobacterium and decreasing the Helicobacter level. Furthermore, liver metabolomic profiling suggested GA intervention had a remarkable regulatory effect on liver metabolism with excessive alcohol consumption. Moreover, GA intervention regulated mRNA levels of alcohol metabolism, fatty lipid metabolism, oxidative stress, bile acid biosynthesis and metabolism-related genes in the liver. Conclusively, these findings demonstrate GA intervention can significantly relieve alcoholic liver injury and it is hopeful to become a new functional food ingredient for the prevention of alcoholic liver injury.
