Ultrasensitive photoelectrochemical detection of glutathione based on the multifunctional catalytic properties of phosphotungstic acid SCIE

Abstract :

A novel photoelectrochemical (PEC) sensor was constructed for the highly sensitive detection of reduced glutathione (GSH) based on the multiple catalytic properties of phosphotungstic acid (PTA). In this work, the catalytic properties of PTA were applied to PEC sensing for the first time and interpreted in detail. First, PTA as an electron acceptor can inhibit the complexation of photogenerated electron-hole pairs in p-Cu2O, thus significantly increasing the photogenerated current of p-type semiconductor material Cu2O. Secondly, when GSH is oxidized to oxidized glutathione (GSSG) by photogenerated holes on the photocathode, PTA is able to reduce GSSG to GSH by transferring protons, forming a redox cycle regeneration process of GSH. Finally, the relatively large amount of PTA in the background solution was able to pre-oxidize interfering substances such as l-cysteine and ascorbic acid, which improved the selectivity of the method. Under the optimal experimental conditions, the linear range of the PEC sensor response to GSH was 0.050-100 nmol L-1, with a detection limit as low as 0.017 nmol L-1 (S/N = 3), which can be applied to the detection of GSH content in cell lysate samples.

Abstract :

In this work, a novel photo-chemiluminescence (PCL) array imaging technique was developed to detect HIV DNA sequences using water-dispersed ultrathin sulfur-doped g-C3N4 porous nanosheets (SCNNSs) as photocatalysts, with complementary chains of HIV DNA as the biorecognition elements. The PCL response was enhanced when a suitable amount of SCNNSs was used. The large specific surface area and p-conjugated structure of the SCNNSs provided a good platform for immobilizing the complementary chains of HIV DNA. When DNA complementary chains were present, some of the catalytically active sites of SCNNSs were blocked, and the PCL of the platform was weakened. When the HIV DNA was added, the DNA double chain was far away from the surfaces of the SCNNSs because p-p the stacking interactions between the formed dsDNA and SCNNSs were weak. Therefore, the addition of the target HIV DNA sequence noticeably restored the signal. In the range of 5.00 x 10(-8) M to 200 x 10(-8) M, the enhanced PCL response was linearly related to the concentration of the HIV DNA sequence, and the detection limit (3S/N) was 1.50 x 10(-8) mol L-1. In addition, the combination of SCNNSs with complementary chains of HIV DNA successfully produced a high-performance PCL imaging sensor. In these proof-of-concept experiments, we demonstrated that our method was fast, portable, and ultra-sensitive, with high throughput.

Abstract :

In recent years, the development of miniature and portable sensors has been a major focus of research. PEC self powered sensors have emerged as a potential solution to the power supply issue, eliminating the need for external power supplies and operating without bias voltage. This study developed a ZnO/Au/GaN sensor for highly sensitive detection of alpha-fetoprotein (AFP). The sensor uses GaN substrates with nanogold films to provide an auxiliary bias voltage, promoting high photogenerated current density. Using ZnO/Au/GaN as a photoanode resulted in significantly higher photocurrent generated by the sensor compared to Au/GaN or ZnO/ ITO alone. To enable selective detection of AFP, antibody modification of the ZnO nanorod arrays was employed. The linear range of the sensor response to AFP was determined to be 0.080-5.0 ng/mL, with an impressively low detection limit of 0.027 ng/mL (S/N = 3). These results demonstrate the potential of this self-powered sensor for detecting AFP content in human serum samples. Overall, this study presents a novel approach for developing highly sensitive and selective self-powered sensors for biomarker detection, which could facilitate early detection and clinical diagnosis of various types of cancer.

Abstract :

The detection of blood glucose plays a crucial role in clinical practice, and the innovative use of sweat as a substitute for blood has brought significant practical value in the detection of blood glucose. However, lactic acid (LA) has been found to be the main interfering substance on the detection of glucose in sweat. In this study, a photoelectrochemical sensing system was constructed by using NiO nanosheets as photosensitive materials and molybdenum-based polyoxometalate clusters (Mo-POM) as masking agents. Mo-POM, an ideal electroncapturing agent, effectively promoted the spatial separation of photo-generated charge carriers in NiO nanosheets, thereby enhancing the oxidation rate of glucose on the valence band of NiO and significantly improving the detection sensitivity. Mo-POM also exhibited the characteristics of an electron sponge, facilitating fast and reversible multi-electron transfer reactions and effectively masking certain redox substances, especially lactic acid, thus greatly enhancing the selectivity of the glucose sensor. The photoelectrochemical (PEC) sensor constructed using Mo-POM has excellent sensing capability with high sensitivity to detect glucose at a minimum concentration of 1 nM. The detection limit of the sensor is 0.33 nM (S/N = 3). This sensor can be reliably applied to the detection of glucose in sweat and the accuracy of the results was verified by gas chromatography-mass spectrometry.

Abstract :

In this work, an Au/GaN photoelectrode was prepared by sputtering a 30.0-nm–thick gold film on an n-type gallium nitride (GaN) surface. Guanine (G) was used as a hole trap in photoinduced charge separation due to its lowest oxidation potential and because it could be easily oxidized among the five common nucleobases. Guanosine-rich DNA sequences can form stable secondary structures known as G-quadruplexes. The efficiency of hole transport was significantly improved after G-quadruplexes formation. The thrombin aptamer rich in guanine could be modified on the electrode surface through Au–S bonds, forming stable G-quadruplexes in the presence of target thrombin. Due to the presence of the G-quadruplexes, the electron-hole recombination rate of the system decreased and the photocurrent increased. Under optimized conditions, high specificity, selectivity, and linearity could be achieved in the concentration range of 0.100–10.0 nm, and the limit of detection was approximately 20.0 pM. Thus, the detection results of human serum thrombin were satisfactory. © 2022 The Authors

Abstract :

MicroRNAs (miRNAs) have been identified as important biomarkers with great significance for diagnosis and treatment of various diseases. However, their unique properties, such as small size, high sequence homology, and low abundance, make quantitative analysis of miRNAs extremely challenging. Herein, we reported a cascade catalytic hairpin assembly (CCHA) for sensitive and selective detection of miRNA with three kinds of hairpin probes (HP1, HP2, and HP3). In the presence of target miRNA, a series of toehold-mediated intermolecular DNA strand displacement and hybridization was activated among HP1, HP2, and HP3 to assembly numbers of DNA nanoobjects. During this period, the fluorescence response was greatly intensified to indicate the presence and expression level of interested target miRNA. We have demonstrated that the proposed method exhibits a high assay sensitivity to detect low concentration target and an excellent sequence specificity to distinguish even a single-nucleotide difference in vitro. Moreover, we also demonstrated that our design enables the intracellular imaging of miRNA in live cancer and normal cells. These results showing the promising potential of our CCHA for powerful biosensing, clinic diagnosis, or prognosis.

Murraya paniculata (L.) is a traditional Chinese medicine (TCM) wildly grown in southeast China, and used for abortion in folk. Murrayone, a coumarin-containing compound extracted from M. paniculata, is the most bioactive substance in this species and is being developed as a novel cancer metastasis chemopreventive agent based on its unique pharmacological properties. In the present study, a novel rapid and sensitive method for quantitative analysis of murrayone in rat plasma and for determining its pharmacokinetics in rats was developed and validated using UPLC/MS/MS. Plasma samples were subjected to protein precipitation and then directly analyzed by UPLC/MS/MS. Both murrayone and coumarin as an internal standard (I.S.) were carried on a C-18 column with a gradient mobile phase consisting of acetonitrile and water at a flow rate of 0.3 mL/min. Several gradient elution procedures were evaluated to achieve effective chromatography resolution and a sensitive response to murrayone and the I.S.. Mass spectrometry was carried out using a triple-quadrupole system via positive electrospray ionization and multiple reaction monitoring (MRM). Good linearity (r(2) = 0.9987) was achieved over a linear range of 4.0-1600 ng/mL with a lower limit of quantitation (LLOQ) of 4.0 ng/mL for murrayone. The inter- and intraday accuracy and precision ranged from 90.0 to 99.7% and 1.1 to 12.3% at four quality control concentrations, respectively. The average absolute recoveries of murrayone and the I.S. were determined to be 85.9-92.4% and 86.5-90.7%, respectively, at 10.0, 80.0, and 800 ng/mL. Murrayone was stable under a variety of storage and processing conditions that may be routinely encountered in laboratories based on all the stability tests. This newly developed method was successfully applied to the pharmacokinetic study of murrayone in rats for the first time, and the current assay methodology could provide important insights into potential therapeutics and facilitate further pharmacodynamic explorations of murrayone. MicroRNAs (miRNAs) are involved in a variety of biological processes, and the accurate detection of miRNAs is of great importance for early diagnosis of various cancers. Herein, we have developed a highly sensitive method for the intracellular imaging of miRNAs based on a palindromic probe-induced strand displacement amplification (pSDA). The sensing element is a partly complementary hybrid consisting of two DNA components: one fluorescent dye-labeled signaling probe containing a palindromic sequence and loop-based target recognition site and one quencher moiety-attached locking probe. In the presence of target miRNA, the target species can hybridize with the loop site and release the terminal palindromic fragment, initiating the pSDA reaction. Thus, a considerable amount of fluorescent moieties are spatially separated from the quenchers, generating a dramatically enhanced fluorescence signal. As a result, the target miRNAs can be quantified down to 25 pM with the linear response range over four orders of magnitude. The detection specificity is high enough to eliminate the interference from nontarget miRNAs and other biospecies co-existing in samples, and thus the diseased cells are easily distinguished from healthy cells. Strikingly, the pSDA-based system possesses the desirable capability to discriminate tumor cells from healthy cells, indicating a promising diagnostic tool for the detection of cancers and other diseases in early stage.
