Abstract :
Photothermal therapy (PTT) has attracted extensive attention in cancer treatment. Heptamethine cyanine dyes with near-infrared (NIR) absorption performance have been investigated for PTT. However, they are often accompanied by poor photostability, suboptimal photothermal conversion and limited therapeutic efficacy. The photophysical properties of fluorescent organic salts can be tuned through counterion pairing. However, whether the counterion can influence the photostability and photothermal properties of heptamethine cyanine salts has not been clarified. In this work, we investigated the effects of eleven counter anions on the physical and photothermal properties of NIR-II heptamethine cyanine salts with the same heptamethine cyanine cation. The anions have great impacts on the physiochemical properties of dyes in solution including aggregation, photostability and photothermal conversion efficiency. The physical tuning enables the control over the cytotoxicity and phototoxicity of the dyes. The selected salts have been demonstrated to significantly suppress 4T1 breast tumor growth with low toxicity. The findings that the counterion has great effects on the photothermal properties of cationic NIR-II heptamethine cyanine dyes will provide a reference for the preparation of improved photothermal agents through counterion pairing with possible translation to humans.

Abstract :
Non-invasive therapies such as photodynamic therapy (PDT) and chemodynamic therapy (CDT) have received wide attention due to their low toxicity and side effects, but their efficacy is limited by the tumor microenvironment (TME), and monotherapy cannot achieve satisfactory efficacy. In this work, a multifunctional nanoparticle co -assembled from oleanolic acid (OA), chlorin e6 (Ce6) and hemin was developed. The as -constructed nanoparticle named OCH with diameters of around 130 nm possessed good biostability, pH/GSH dual -responsive drug release properties, and remarkable cellular internalization and tumor accumulation capabilities. OCH exhibited prominent catalytic activities to generate center dot OH, deplete GSH, and produce O 2 to overcome the hypoxia TME, thus potentiating the photodynamic and chemodynamic effect. In addition, OCH can induce the occurrence of ferroptosis in both ferroptosis-sensitive and ferroptosis-resistant cancer cells. The multi -pronged effects of OCH including hypoxia alleviation, GSH depletion, ferroptosis induction, CDT and PDT effects jointly facilitate excellent anticancer effects in vitro and in vivo . Hence, this work will advance the development of safe and effective clinically transformable nanomedicine by employing clinically -applied agents to form drug combinations for efficient multi -pronged combination cancer therapy.

Abstract :
Inorganic antimicrobial materials have the advantages of high safety, strong durability, stable antimicrobial performance, and good heat resistance compared with organic antimicrobial materials. In this study, Fe-doped small zinc oxide nanoparticles (FZO-NPs) with different iron contents were synthesized and then modified with 3-aminopropyltriethoxysilane (APTES) and poly(ethylene glycol)-600 (PEG-600) to afford FZO-APs. FZOAPs which were fully characterized can disperse in water with good dispersity and improved stability. In antimicrobial tests, FZO-AP4 displayed excellent antimicrobial effects under 10 min of 365 nm UV light against Gram-negative bacteria Escherichia coli (E. coli) and Gram-positive bacteria Staphylococcus aureus (S. aureus) with a minimum inhibitory concentration (MIC) of 0.20 and 0.15 mg/mL, respectively. The antimicrobial mechanism studies revealed that the consumption of glutathione (GSH) and ascorbic acid (ASA), and the generation of reactive oxygen species (ROS) could trigger lipid peroxidation (LPO) and ultimately induce ferroptosis in bacterial cells. The cause of the cell wall/membrane damage and the released Zn/Fe ions from the nanoparticles inside the bacterial cells further promote the antimicrobial effects. In addition, FZO-AP4 showed high inhibitory effects on bacterial biofilm formation and significantly disrupted the formed biofilm. Interestingly, higher bacterial biofilm residuals and bacterial survival were found in E. coli than in S. aureus after FZO-AP4 treatment, suggesting a potential link between the quorum sensing (QS) system of the bacteria and their different susceptibility towards FZO-AP4. This study proved the antibacterial effects of small water-soluble FZO-APs which is meaningful for the design and development of new inorganic nano-material for antimicrobial applications.

Abstract :
Photothermal therapy (PTT) has attracted extensive attention in cancer treatment because of its advantages of non-invasive and few side effects. Gold nanoparticles and boron-fluorine-dipyrrole (BODIPY) dye have been used as photothermal agents (PTAs), but they have problems such as easy aggregation, poor water solubility and low photothermal conversion efficiency. In this work, we used conventional liposomes for in situ generation of small gold nanoparticles and sequentially loading BODIPY to overcome the drawbacks of the two PTAs and simultaneously realize enhanced PTT. Gold- and BODIPY-coordinated liposomal nanocomplexes LAB with small gold nanoparticles adsorbed on the surface and BODIPY entrapped inside showed uniform size of about 100 nm with good stability. LAB demonstrated laser-responsive drug release property, good cellular uptake efficiency and tumor accumulation ability. After laser irradiation, LAB can exert enhanced PTT in cancer cell lines by reducing mitochondrial membrane potential and increase intracellular reactive oxygen species level to promote cell apoptosis. In tumor-bearing mice, LAB showed fantastic tumor inhibition effect with good biosafety. This work provides a convenient method to develop PTA combinations for enhanced PTT, which could lay the foundation for the green construction of new PTAs for PTT by employing clinically used formulations and PTAs.

Therapy-induced modulation of the tumor microenvironment (TME) to overcome the immunosuppressive TME is considered to be an opportunity for cancer treatment. However, monitoring of TME modulation during the therapeutic process to accurately determine immune responses and adjust treatment plans in a timely manner remains to be challenging. Herein, we report a carrier-free nanotheranostic system (CANPs) assembled by two boron dipyrromethene (BODIPY) dyes, a sonophotosensitizer C-BDP, and a nitric oxide (NO) probe amino-BODIPY (A-BDP). CANPs can exert combined sonophototherapeutic effects of C-BDP under ultrasound and light irradiation and simultaneously induce inflammatory TME, as well as emit bright fluorescence via A-BDP by monitoring tumor-associated macrophages (TAMs) repolarization through the released NO in vitro and in vivo. Of note, transforming growth factor-beta (TGF-beta) could be the key cytokine involved in the sonophototherapy-induced TME reprogramming. By virtue of high physiological stability, good biocompatibility, and effective tumor targetability, CANPs could be a potential nanotheranostic system for the simultaneous induction and detection of TME reprogramming triggered by sonophototherapy.

The therapeutic effectiveness of photothermal therapy (PTT) is limited by heat tolerance and PTT-induced inflammation, which increases the risk of tumor metastasis and recurrence. Ferroptosis combined with PTT can achieve significant therapeutic effects. In this work, we designed self-healing photothermal nanotherapeutics to achieve effective PTT with triple-amplified ferroptosis and cascade inflammation inhibition after photothermal treatment. After the ferroptosis-inducing ability of mangiferin (MF) was first elucidated, the nanocomplex PFeM, coordinated by Fe3+ and MF with polyvinylpyrrolidone (PVP) modification, was prepared by a one-pot self-assembly method. PFeM with laser irradiation could induce intensified ferroptosis by integrating the functions of MF to deactivate glutathione peroxidase 4, Fe3+/Fe2+ to generate lethal reactive oxygen species via the Fenton reaction, and the photothermal effect to amplify ferroptosis. More importantly, the released MF could achieve cascade inflammation inhibition, thereby reversing the proinflammatory microenvironment caused by PTT. The in vivo antitumor and anti-inflammatory effects of PFeM were further confirmed in a 4T1 tumor-bearing mouse model. This study expounding the ferroptosis-inducing effects of MF and utilizing the strategy of chelating MF with iron ions can provide a new idea for developing photothermal nanoagents with clinically convertible safety ingredients and a green preparation process that improve efficacy and reduce adverse reactions during PTT. © 2024 American Chemical Society.

Abstract :
The emergence of acquired resistance to epidermal growth factor receptor tyrosine kinase inhibitors (EGFR-TKIs) poses formidable obstacles to non-small cell lung cancer (NSCLC) therapy. Real-time monitoring of the efficacy of EGFR-TKIs to avoid invalid treatment remains a great challenge. Herein, we rationally designed a EGFR-TKImodified liposomal complex co-loaded with a iodine-containing contrast agent ioversol (IOV) and a photosensitizer indocyanine green (ICG), namely ELIG, for simultaneous EGFR-TKI-mediated computed tomography/ fluorescence dual-modal imaging to monitor drug resistance during the treatment, and combined photodynamic therapy (PDT) and photothermal therapy (PTT) to reverse drug resistance. ELIG exhibited good physicochemical properties, favorable photothermal effects, and high singlet oxygen production. ELIG can protect ICG and IOV from rapid degradation and prolong the circulation time. In addition, ELIG had a great ability to recognize drugsensitive EGFR-mutated NSCLC cells, and could monitor the changes in the intracellular amounts of the imaging agents to detect the drug resistance and predict the efficacy during the treatments. Additionally, the heavy atom effect of IOV can increase the PDT effect of ICG obviously. With 808 nm laser irradiation, ELIG showed satisfactory anticancer effects to reverse drug resistance by the synergism of PDT and PTT in vitro and in vivo. Hence, this work holds great promise for providing a new idea and an effective strategy for predicting and managing the efficacy of EGFR-TKIs.

Herein, we report an efficient approach to synthesize artemisinin G from artemisinin via photocatalysis in high yield. Herein, an efficient method for the synthesis of artemisinin G from artemisinin by using 0.5 mol % of TBADT as the catalyst was reported. Inducing redox imbalance in cancer cells aided by external stimuli is a promising strategy to treat cancer. However, maximizing efficacious reactive species content with durable effects in deep tumors remains to be challenging. Here, we report an herbal medicine-based nanoreactor (OH7) self-assembled by oleanolic acid (OA) and two sonosensitizers. OH7 couples two cascade reactions under sonication, including dual-sonosensitizer-participated consecutive reactions for generating long-living peroxyl radical (ROO center dot) and OA-participated se-rial reactions for forming highly cytotoxic peroxynitrite (ONOO-). The site-directed reactive species burst exerts excellent cytotoxicity on two-dimensional cancer cell cultures and multicellular tumor spheroids. The high elasticity of OH7 for deep tumor transportation, the enhanced intratumoral convection by ultrasound, and the formation of ONOO- to destroy extracellular matrix jointly facilitate satisfactory penetration depth of sono-sensitizers. Of note, the deep penetration and the long-lived hypertoxic reactive species generating ability can realize effective tumor inhibition by systemic administration. This study opens up new insights into the reactive species-related bioactive mechanisms to support green development of biocompatible high-performance sono-catalytic nanoreactor for potential clinical applications.

Abstract :
Tetrahydro-beta-carbolines (TH beta Cs) are a kind of natural alkaloids with multiple pharmaceutical activities. Herein, a focused compound library derived from TH beta Cs was synthesized and their anticancer activities were studied in several cancer cell lines. Among them, three compounds showed considerable anticancer activities with low micromolar to submicromolar IC50 values. The abilities to induce apoptosis and alter mitochondrial membrane potential levels, which are comparable to those of the commercial anticancer drug adriamycin, were confirmed by one representative compound (21) on the B16/F10 cell line. Our preliminary structure-activity relationship studies indicated that alkylamines with suitable lengths are very important for potency improvement.
