A Drosophila Model Reveals the Potential Role for mtt in Retinal Disease SCIE

Abstract :

Congenital stationary night blindness (CSNB) is a genetically heterogeneous inherited retinal disorder, caused by over 300 mutations in 17 different genes. While there are numerous fly models available for simulating ocular diseases, most are focused on mimicking retinitis pigmentosa (RP), with animal models specifically addressing CSNB limited to mammals. Here, we present a CSNB fly model associated with the mtt gene, utilizing RNA interference (RNAi) to silence the mtt gene in fly eyes (homologous to the mammalian GRM6 gene) and construct a CSNB model. Through this approach, we observed significant defects in the eye structure and function upon reducing mtt expression in fly eyes. This manifested as disruptions in the compound eye lens structure and reduced sensitivity to light responses. These results suggest a critical role for mtt in the function of fly adult eyes. Interestingly, we found that the mtt gene is not expressed in the photoreceptor neurons of adult flies but is localized to the inner lamina neurons. In summary, these results underscore the crucial involvement of mtt in fly retinal function, providing a framework for understanding the pathogenic mechanisms of CSNB and facilitating research into potential therapeutic interventions.

Abstract :

The effects of packaging and storage conditions on the moisture content and instrumental and sensory textural properties of raw and salty light roasted (SLR) California almonds were studied under different storage conditions. The controlled combinations included low, medium, and high temperatures and low and high relative humidity (RH). Almond samples were packaged in cartons or polyethylene (PE) bags with and without vacuum. Both absorption and desorption of moisture by almonds were observed during storage and were dependent on the packaging and storage conditions. In general, gradual changes were observed for samples with PE and vacuum PE packaging in most of the storage conditions, while the samples packed in cartons showed more dramatic changes because these unprotected samples were more vulnerable to seasonal changes in humidity. The SLR almonds showed consistent moisture gains, while the raw almonds tended to lose moisture content in most of the storage conditions. This may be attributed to the low initial moisture content of the SLR samples. All raw samples packed in cartons became softer over time. The softening tended to be enhanced by high storage humidity and temperature. The raw almonds packaged in PE bags were firmer than those packed in cartons but also became softer over time. The firmness of the SLR samples was generally lower than that of raw samples, probably because roasting reduced the density and mechanical strength of the kernels. The firmness of PE packaged SLR samples increased in uncontrolled storage conditions and in higher storage temperature and humidity conditions but decreased slightly in lower temperature and humidity conditions. Vacuum packaging did not affect the firmness much. Using PE packaging and maintaining the RH below 50% and the temperature below 25 degrees C are effective in stabilizing both raw and processed almonds.

Long non-coding RNAs regulate Wnt signaling during feather regeneration SCIE

Abstract :

Long non-coding RNAs (lncRNAs) are non-protein coding transcripts that are involved in a broad range of biological processes. Here, we examine the functional role of lncRNAs in feather regeneration. RNA-seq profiling of the regenerating feather blastema revealed that Wnt signaling is among the most active pathways during feather regeneration, with Wnt ligands and their inhibitors showing distinct expression patterns. Co-expression analysis identified hundreds of lncRNAs with similar expression patterns to either the Wnt ligands (the Lwnt group) or their downstream target genes (the Twnt group). Among these, we randomly picked two lncRNAs in the Lwnt group and three lncRNAs in the Twnt group to validate their expression and function. Members in the Twnt group regulated feather regeneration and axis formation, whereas members in the Lwnt group showed no obvious phenotype. Further analysis confirmed that the three Twnt group members inhibit Wnt signal transduction and, at the same time, are downstream target genes of this pathway. Our results suggest that the feather regeneration model can be utilized to systematically annotate the functions of lncRNAs in the chicken genome.

We report the synthesis of two-dimensional Te nanosheets through a facile liquid exfoliation method. The as-synthesized Te nanosheets can produce reactive oxygen species under light irradiation and show high photoacoustic imaging performance due to their strong near-infrared absorbance, and can be engineered as a nanoplatform for simultaneous photoacoustic imaging and photodynamic therapy.

Abstract :

The objective of this study was to investigate the fundamental question of biofilm formation. First, a drum biofilm reactor was introduced. The drums were coated with three porous substrates (cotton rope, canvas, and spandex), respectively. The relationships among the substrate, extracellular polymeric substances (EPS), and adhesion ratio were analyzed. Second, a plate biofilm reactor (PBR) was applied by replacing the drum with multiple parallel vertical plates to increase the surface area. The plates were coated with porous substrates on each side, and the nutrients were delivered to the cells by diffusion. The influence of nitrogen source and concentration on compositions of EPS and biofilm formation was analyzed using PBR under sunlight. The results indicated that both substrate and nitrogen were critical on the EPS compositions and biofilm formation. Under the optimal condition (glycine with concentration of 1 g l(-1) and substrate of canvas), the maximum biofilm productivity of 54.46 g m(-2) d(-1) with adhesion ratio of 84.4 % was achieved.

The microorganisms in biofilms live in a self-produced matrix of hydrated extracellular polymeric substances (EPS) that forms their immediate environment. The objective of this paper was to investigate the relationships between culture conditions, EPS, microalgal biofilms growth and lipid accumulation. Fresh water alga Botryococcus braunii was attached culture in multi-layers photobioreactors with different culture media and substrates. The results indicated that the production of EPS was affected by culture period, nutrient, and substrate. Increasing the production of EPS may enhance the biofilm growth. However, the EPS components, namely proteins and polysaccharide had a more profound effect on biofilm formation compared to total EPS, with protein being more significant than polysaccharide. Nitrogen-free and EPS strategies were conducted to increase the lipid content of B. braunii biofilm from 11.6 to 42.3 % and 51.3 %, respectively. Compared to suspended culture, the lipid quality was enhanced. The dominant component of hexadecanoic acid (16:0) was enhanced from 21.78 to 48.17 % and 55.44 %, respectively.

Dissecting the Molecular Mechanism of Ionizing Radiation-Induced Tissue Damage in the Feather Follicle SCIE

Abstract :

Ionizing radiation (IR) is a common therapeutic agent in cancer therapy. It damages normal tissue and causes side effects including dermatitis and mucositis. Here we use the feather follicle as a model to investigate the mechanism of IR-induced tissue damage, because any perturbation of feather growth will be clearly recorded in its regular yet complex morphology. We find that IR induces defects in feather formation in a dose-dependent manner. No abnormality was observed at 5 Gy. A transient, reversible perturbation of feather growth was induced at 10 Gy, leading to defects in the feather structure. This perturbation became irreversible at 20 Gy. Molecular and cellular analysis revealed P53 activation, DNA damage and repair, cell cycle arrest and apoptosis in the pathobiology. IR also induces patterning defects in feather formation, with disrupted branching morphogenesis. This perturbation is mediated by cytokine production and Stat1 activation, as manipulation of cytokine levels or ectopic Stat1 over-expression also led to irregular feather branching. Furthermore, AG-490, a chemical inhibitor of Stat1 signaling, can partially rescue IR-induced tissue damage. Our results suggest that the feather follicle could serve as a useful model to address the in vivo impact of the many mechanisms of IR-induced tissue damage.

Avian feathers have robust growth and regeneration capability. To evaluate the contribution of signaling molecules and pathways in these processes, we profiled gene expression in the feather follicle using an absolute quantification approach. We identified hundreds of genes that mark specific components of the feather follicle: the dermal papillae (DP) which controls feather regeneration and axis formation, the pulp mesenchyme (Pp) which is derived from DP cells and nourishes the feather follicle, and the ramogenic zone epithelium (Erz) where a feather starts to branch. The feather DP is enriched in BMP/TGF-beta signaling molecules and inhibitors for Wnt signaling including Dkk2/Frzb. Wnt ligands are mainly expressed in the feather epithelium and pulp. We find that while Wnt signaling is required for the maintenance of DP marker gene expression and feather regeneration, excessive Wnt signaling delays regeneration and reduces pulp formation. Manipulating Dkk2/Frzb expression by lentiviral-mediated overexpression, shRNA-knockdown, or by antibody neutralization resulted in dual feather axes formation. Our results suggest that the Wnt signaling in the proximal feather follicle is fine-tuned to accommodate feather regeneration and axis formation. (C) 2014 Elsevier Inc. All rights reserved.

The objective of this study was to understand and optimize the formation of microalgae biofilms in specific culture conditions. Firstly, the adhesion of six freshwater algae species was compared. Chlorococcum sp. was selected because of the high adhesion biomass productivity (ABP) and adhesion rate achieved. Secondly, the adhesion of Chlorococcum sp. was compared with nine commonly used supporting materials, and glass fiber-reinforced plastic proved to be the optimal substrata. Thirdly, based on response surface methodology experiments, a second-order polynomial model was developed to examine the effect of culture period, initial total nitrogen concentration (ITNC) in manure wastewater, pH and culture volume of the growth chamber on the adhesion of Chlorococcum sp. using glass fiber-reinforced plastic. The experimental and modeling results showed that ITNC, pH and culture volume as well as the interactions between culture period and ITNC, culture period and culture volume were significant on ABP. Optimum culture conditions were predicted at a culture period of 11 days, ITNC of 70 mg L-1, pH of 8 and culture volume of 340 mL, under which the predicted maximum ABP was 4.26 g m(-2) day(-1). The prediction was close to validation experimental results, indicating that the model could be used to guide and optimize the attached culture of Chlorococcum sp. using glass fiber-reinforced plastic.

Three New Oblongolides from Phomopsis sp XZ-01, an Endophytic Fungus from Camptotheca acuminate SCIE

Four new metabolites, including three new oblongolides named C1, P1, and X1 (1-3) and 6-hydroxyphomodiol (10), along with eight known compounds - oblongolides B (4), C (5), D (6), O (7), P (8) and U (9), (3R,4aR,5S,6R)-6-hydroxy-5-methylramulosin (11), and (3R)-5-methylmellein (12) - were isolated from the endophytic fungal strain Phomopsis sp. XZ-01 of Camptotheca acuminate. Their structures were elucidated by spectroscopic analyses, including H-1- and C-13-NMR, 2D NMR (HSQC, HMBC, H-1-H-1 COSY and NOESY) and HR-FT-MS. Cytotoxic activities of these compounds were evaluated. Some of them showed weak selective activities.
