Abstract :

Nanozymes are ideal materials for designing colorimetric sensors, and the spontaneous catalytic effect in most of nanozymes commonly leads to instability of the colorimetric signal. To improve this limitation, photoresponsive nanozymes as a switch under specific-light irradiation are used to regulate the reaction process in a controlled manner. Strontium titanate (SrTiO3) is a high-performance photocatalyst, whose heterojunctions formed with reduced graphene oxide (rGO) are promising to efficiently induce the separation of carriers, further generating abundant radicals to colorize the substrate. Here, the SrTiO3-rGO composites with a high specific surface area were synthesized by hydrothermal method and showed excellent peroxidase-like activity under ultraviolet irradiation. The affinity for 3,3',5,5'-tetramethylbenzidine is equivalent to 19 times that of horseradish peroxidase and the reaction mechanism is analyzed based on reactive intermediates(center dot O-2(-)) and elemental valence changes. A colorimetric sensor based on SrTiO3-rGO with high sensitivity, low limit (56 nM), and wide linear range (1-100 mu M) is constructed for detecting tannins in tea samples. This visualization model provides a reliable idea for the quantitative analysis of trace substances.

Abstract :

Photodynamic therapy (PDT), as an alternative non-invasive clinical treatment modality, has been extensively employed for various anticancer applications in preclinical and clinical trials. However, there is hardly any PDT research based on invasive or metastatic tumors. But it can't be ignored that cancer metastases are responsible for 90% of all cancer-related death. Among these metastatic cancers, prostate cancer (PCa) is regarded as one of the leading causes of cancer related deaths amongst male patients due to its high metastasis and high fatality rate. At present, it remains a great challenge to apply traditional PDT technology for inhibiting prostate cancer metastasis. In this work, we successfully designed and synthesized a novel MAO-A targeted photosensitizer Pc-MLB for targeted photodynamic treatment of prostate cancer and inhibiting its metastasis in vitro. In vitro ex-periments demonstrate that Pc-MLB shows high target affinity and specificity towards prostate cancer cells and remarkable photodynamic anticancer activity compared with the control. More significantly, the migration and invasion assays show that Pc-MLB exhibits the ability to inhibit the migration and metastasis of prostate cancer cells to some extent.

Abstract :

Photodynamic therapy (PDT) is a promising and non-invasive treatment for various cancers. Although nuclear PDT has considerable therapeutic prospects, it is still hindered by the non-specific recognition of tumor tissues or the degradation of nuclear targeting cationic groups by enzymes in the blood. Herein, a hierarchical targeted and controlled release strategy is proposed by using folate-modified poly-beta-cyclodextrin (poly-beta-CD) as a nano-carrier for loading nuclear localization signals (NLSs)-conjugated photosensitizer PAP (PAP = pyropheophorbide a-PAAKRVKLD). Excitingly, the obtained FA-CD@PAP (FA = folic acid) and nanoparticles (NPs) can specifically recognize tumor cells overexpressing folate receptors (FR) to remarkedly enhance the intracellular accumulation. Furthermore, the encapsulated PAP can be released under acidic conditions to realize precise nuclear localization. The reactive oxygen species (ROS) generated by the intranuclear-accumulated PAP upon irradiation can oxidize and destroy DNA chains or DNA repair enzymes instantaneously, which can directly induce cell death. As a result, FA-CD@PAP NPs exhibit excellent tumor regression and negligible side effects. This work provides an intelligent nuclear-targeted delivery strategy for in situ nuclear PDT with extremely prominent efficacy and high biological safety.

Abstract :

Multi-modal synergistic therapy, especially the integration of near-infrared laser phototherapies and chemo-therapy, is often sought after owing to its minimal invasiveness, low side effects, and improved anticancer therapeutic efficacy. Herein, CuS nanoparticles were first coated with zinc phthalocya-nine derivant (Pc)-functionalized mesoporous silica (mSiO2-Pc) to achieve a drug delivery system (CuS@mSiO2-Pc) with photo-thermal/photodynamic therapy. Chemical drug DOX was subsequently loaded for chemotherapy, and hyaluronic acid (HA) was employed as a covering material with cancer targeting. The as-obtained CuS@mSiO2-Pc(DOX)@HA nanoparticles were nano-sized with good biocompatibility, effective DOX loading, and controllable DOX releasing. Expectedly, this multifunctional nanoplatform exhibits effective generation of reactive oxygen species and hyperthermia upon the near-infrared laser irradiation. Most importantly, the nanoparticles were targeted into 4T1 cells and showed significantly remarkable cytotoxicity under near -infrared laser irradiation, proving their synergistic therapeutic efficacy. Therefore, this targeted drug system based on CuS with synergistic photothermal therapy/photodynamic therapy/chemotherapy has great application prospects in clinical anticancer treatment for triple negative breast cancer.

Abstract :

Tumor acidic environment-activated combination therapy holds great promise to significantly decrease side effects, circumvent multiple drug resistance, and improve therapeutic outcomes for cancer treatment. Herein, Sorafenib/ZnPc(PS)(4)@Fe-III-TA nanoparticles (SPFT) are designed with acid-environment turned-on fluorescence to report the activation of triple therapy including photodynamic, chemodynamic, and chemotherapy on hepatocellular carcinoma. The SPFT are composed of SP cores formulated via self-assembly of sorafenib and ZnPc(PS)(4), with high drug loading efficiency, and Fe-III-TA shells containing FeCl3 and tannic acid. Importantly, the nanoparticles suppress reactive oxygen species (ROS) generation of ZnPc(PS)(4) due to their formation in nanoparticles, while assisting simultaneous uptake of the uploaded drugs in cancer cells. The tumor acidic environment initiates Fe-III-TA decomposition and accelerates a chemodynamic reaction between Fe(II )and H2O2 to generate toxic center dot OH. Then, the SP core is decomposed to separate ZnPc(PS)(4) and sorafenib, which leads to fluorescence turning-on of ZnPc(PS)(4), expedited photodynamic reactions, and burst release of sorafenib. Notably, SPFT shows low dark cytotoxicity to normal cells but exerts high potency on hepatocellular carcinoma cells under near-infrared light irradiation, which is much more potent than either sorafenib or ZnPc(PS)(4) alone. This research offers a facile nanomedicine design strategy for cancer therapy.

Abstract :

A detailed study of topological phase transition is essential for guiding the controlled synthesis of materials. Herein, a series of zeolitic-imidazolate frameworks (ZIFs) and their derived metal oxides with tailored morphologies and adjustable phases from ZIF-L to ZIF-8 to ZnO were reported. More significantly, the phase transformation and morphology control of the product could be achieved by simply changing the molar/volume ratio of the reagents without adding any surfactant, and the growth mechanism was systemically investigated. Benefiting from their unique structural advantage, when fabricating gas sensors, all of the products and their derivatives exhibited excellent gas-sensing selectivity and rapid response/recovery times toward acetone vapor. In addition, the gas-sensing mechanism and the causes for the gas-sensing performance discrepancy in different sensors were also studied. The results showed that the oxygen vacancy (O-V) content, structures, and morphologies of the product were closely related to their comparable sensitivities. The phase transition and morphology evolution of ZIFs and ZnO in this study might bring a deep understanding of the metal-organic framework crystallization process, which is expected to help the development of more precise control strategies for MOF-based materials with customized functional properties.

With respect to the probabilistic hesitant fuzzy two-sided matching decision making problems with aspiration levels on criteria, a two-sided matching decision making method based on the regret theory is proposed considering the psychological behaviour of regret aversion of two-sided matching subjects. Firstly, a new Lance distance measure and a Lance score function of the probabilistic hesitant fuzzy element are defined. Then, based on the regret theory, a probabilistic hesitant fuzzy two-sided matching decision making method with aspiration levels on criteria is proposed. In this method, the criterion utility value matrix of two-sided matching subjects are constructed using the expected utility function and the Lance score function of the probabilistic hesitant fuzzy element, the regret-rejoice value matrix of two-sided matching subjects relative to the aspiration levels on criteria are built using the regret-rejoice function, and the perceived value matrix of two-sided matching subjects relative to the aspiration levels on criteria are set up according to the regret theory. Furthermore, an optimization model is constructed using the ideas of maximal deviation method and the new Lance distance measure of the probabilistic hesitant fuzzy element to obtain the weight vectors of the criteria, the comprehensive perceived value matrix of two-sided matching subjects are built using the linear weighted method, and the satisfaction degree matrix of two-sided matching subjects are set up using the range transformation method. Based on these, a multi-objective optimization model is constructed considering maximal satisfaction degree and the stability of two-sided matching alternatives, and the stable two-sided matching outcomes are obtained by solving this model. Finally, an example is given to illustrate the effectiveness and practicability of the proposed two-sided matching decision making method. © 2022 Northeast University. All rights reserved.

Abstract :

In this research, a conjugated Pc-MIL-88B (Fe) nanoplatform was constructed via a condensation process between modified zinc phthalocyanine and MIL-88B (Fe) for the removal of organic pollutants. The as-prepared material was fully characterized by TEM, XPS, ICP, FTIR, UV-Vis, N-2 adsorption-desorption isotherm, etc. The results indicate that Pc-MIL-88B (Fe) preserved the topological structure of MIL-88B (Fe), and the micropores of framework could effectively prevent the aggregation of Pc in water. Meanwhile, the conjugated Pc-MIL-88B (Fe) basically maintains the singlet oxygen quantum yield of Pc, and behaves a much higher photocurrent intensity compared to NH2-MIL-88B (Fe). Additionally, the photosensitive activity and reusability of Pc-MIL-88B (Fe) were evaluated by the degradation of methylene blue in aqueous solution under visible light irradiation, and the degradation mechanism was also investigated in detail.

Harmful bacteria have seriously threatened human health and wealth for a long time. Herein, a multifunctional drug delivery system based on UiO-66 was fabricated, and it showed potent synergistic antibacterial effects when used in conjunction with photodynamic therapy and chemotherapy. First, UiO-66-NH2 was prepared via a facile solvothermal method. Then, carboxylic zinc phthalocyanine, a broad-spectrum photosensitizer, was connected to UiO-66-NH2 by amidation. Next, synergistic chemical antibiotic linezolid was loaded in the pores, and lysozyme was coated on the surface by electrostatic interactions. In vitro antibacterial experiments were then carried out to evaluate the antibacterial effects of this system against three kinds of bacteria, Staphylococcus aureus, Escherichia coli, and methicillin-resistant S. aureus (MRSA). The combination of lysozyme, linezolid, and singlet oxygen generated by irradiation of the photosensitizers resulted in a potent antibacterial effect against S. aureus, E. coli, and even MRSA, which demonstrates the synergistic antibacterial efficacy of photodynamic therapy and chemotherapy.

Dasatinib is a kinase-targeted drug used in the treatment of leukemia. Regrettably, it remains far from optimal medicine due to insurmountable drug resistance and side effects. Photodynamic therapy (PDT) has proven that it can induce systemic immune responses. However, conventional photosensitizers as immunomodulators produce anticancer immunities, which are inadequate to eliminate residual cancer cells. Herein, a novel compound 4 was synthesized and investigated, which introduces dasatinib and zinc(II) phthalocyanine as the targeting and photodynamic moiety, respectively. Compound 4 exhibits a high affinity to CCRF-CEM cells/tumor tissues, which overexpress lymphocyte-specific protein tyrosine kinase (LCK), and preferential elimination from the body. Meanwhile, compound 4 shows excellent photocytotoxicity and tumor regression. Significantly, compound 4-induced PDT can obviously enhance immune responses, resulting in the production of more immune cells. We believe that the proposed manner is a potential strategy for the treatment of T-cell acute lymphoblastic leukemia.
