Abstract :
In view of the biological significance and micro-heterogeneity of protein glycosylation for human health, specific enrichment of N-glycosylated proteins/peptides from complex biological samples is a prerequisite for the discovery of disease biomarkers and clinical diagnosis. In this work, we propose a "grafting-from" N-glycoprotein enriching method based on the in-situ growth of thermoresponsive polymer brushes from the N-glycosylated site of proteins. The initiator was first attached to the pre-oxidized glycan moieties by hydrazide chemistry, from which the thermoresponsive polymers can be grown to form giant protein-polymer conjugates (PPC). The thermosensitive PPC can be precipitated and separated by raising the temperature to above its lower critical solubility temperature (LCST). Mass spectrometry verified 210 N-glycopeptides corresponding to 136 N-glycoproteins in the rabbit serum. These results demonstrate the capability of the tandem thermoprecipitation strategy to enrich and separate N-glycoprotein/glycopeptide. Due to its simplicity and efficiency specifically, this method holds the potential for identifying biomarkers from biological samples in N-glycoproteome analysis.

Abstract :
Bacterial infection of wounds is one of great concern to patients, and rapid and correct detection of bacterial infection is crucial to ensure accurate diagnosis and early intervention. Based on the principle that glucose can only be consumed by live bacteria, a fluorescent biosensor was constructed to detect four kinds of common infectious bacteria (Acinetobacter baumannii, Escherichia coli, Pseudomonas aeruginosa and Staphylococcus aureus) in skin wounds, taking advantage of magnetic bead-aptamer for recognizing, sorting and enrichment, platinum nanoparticles for signal amplification. The linear detection range of AB, EC, PA and SA were 27- 8 x 106 CFU/mL, 10- 2.5 x 107 CFU/mL, 34- 2.5 x 107 CFU/mL and 10- 1.0 x 107 CFU/mL, respectively, and the limits of detection were 27 CFU/mL, 10 CFU/mL, 34 CFU/mL and 2 CFU/mL. Furthermore, all target bacteria in samples containing 8 x 108 CFU/mL of other interfering bacteria have been successfully identified and quantified. The proposed method was also successfully applied to the detection of bacterial infection in skin wounds of mouse and human, including the detection of separate bacterial infection as well as a coinfection. The recovery of this method was in the range of 90.161- 109.961%. Thus, this proposed method can be a promising candidate for rapid and convenient evaluation of infectious bacteria in point-of-care settings.

Abstract :
The combined contamination of multiple mycotoxins (MTs) is a matter of utmost concern, posing significant threats to both the economy stability and the well-being of humans and animals. Consequently, there is an urgent demand for highly sensitive and portable multiplex detection methods to effectively identify and quantify MTs. The primary objective of this paper is to provide a comprehensive overview of the methods to detect multiple MTs in agricultural products, including instrumental and biosensor approaches published in the last five years. Specifically, we emphasize detection methods for antibody (ab)- and aptamer (apt)-based biosensors, while undertaking a comparative analysis of their performance, particularly focusing on the sensitivity. Furthermore, this review proposes several promising technologies that can be leveraged in the future and outlines the challenges and prospects associated with achieving rapid, accurate, and intelligent detection of MTs.

Abstract :
The gelatin (GL) based film with high antioxidative and antibacterial activity and emulsifier-free was developed. The tensile strength (TS) and elongation at break (EAB) of gelatin films could be improved by adding TEMPO oxidized cellulose nanofibers (TCNFs). When the content of epigallocatechin gallate (EGCG) and carvacrol (CAR) reached 20 mg and 500 mg, the EAB of the composite film was 61.25%, which was much higher than that of gelatin film (28.73%). EGCG and CAR could improve UV barrier performance of GL/TCNFs films. GL/TCNFs/ 20EGCG/500CAR film allowed DPPH. and ABTS+ free radical scavenging rates to reach 92.6% and 90.6%, and the diameters of inhibition zones of E. coli and S. aureus were 22.28 mm and 42.58 mm, which showed that EGCG and CAR had synergistic effects. In the coating preservation of sea bass fillets, it also showed excellent inhibition of bacterial proliferation including Pseudomonas aeruginosa, H2S-producing bacteria as well as yeast and mold. TVB-N and TBA of control group were 15.21 mg/100g and 0.353 mg MDA/kg respectively on day 6, while lower values of 5.41 mg/100g and 0.124 mg MDA/kg respectively were recorded for GL/TCNFs/EGCG/500CAR group at the end of storage. All these results indicated that GL/TCNFs/EGCG/CAR film has potential application in fish preservation and is an environmentally friendly active food packaging system.

In this work, silver ion functionalized covalent organic polymer (COPTAPB-BPDA@SA@Ag+) was fabricated via the introduction of silver ion to COP, and utilized as selective sorbent for online SPME of unsaturated fatty acid methyl esters (UFAMEs). The synthesis of COPTAPB-BPDA@SA@Ag+ was confirmed by using the SEM, EDS, FT-IR, XRD and XPS characterization techniques. Due to the charge-transfer interaction between silver ion and C = C double bonds, COPTAPB-BPDA@SA@Ag+ displayed satisfactory extraction ability towards UFAMEs. Several SPME factors, such as ACN content in the sampling solution and eluent, sampling and elution flow rate, were investigated with respect to the extraction efficiency of UFAMEs. Under the optimized conditions, an online Ag+- SPME-Ag+-HPLC method for analysis of target trans-UFAMEs was proposed. A good linearity was established in the range of 0.04 ~ 2 mg/kg for two target trans-UFAMEs in milk tea samples. The limits of detection (LODs) and limits of quantitation (LOQs) were not more than 8.8 mu g/kg and 19 mu g/kg respectively, while the intra-day RSDs (n = 3) and the inter-day RSDs (n = 3) were less than 2.1% and 2.9% respectively. The recoveries for the proposed method was ranged from 82.3% to 93.5% with RSDs (n = 4) less than 5.9%. The above results demonstrated the selective extraction of COPTAPB-BPDA@SA@Ag+ towards UFAMEs from complex real samples, as well as the satisfactory detection sensitivity for trans-fatty acids of the proposed method. This study provided a facile approach for the preparation of metal ion functionalized COP sorbent and showed its wide potential for sample preparation.

In this work, poly(vinylidene fluoride) (PVDF) membranes with hydrophilicity as well as preeminent mechanical strength and dye removal efficiency were fabricated by blending with three dimensional hydroxyapatite nanoparticles (HAPNPs). Surface chemical composition and morphology of the prepared membranes were systematically investigated by ATR-FTIR, XPS, XRD, FESEM, and EDS mapping analyses. The results verified that a large number of HAPNPs were successfully embedded on the modified membrane cross-sections. Moreover, HAPNPs content in the casting solution is an important factor that could have profound influence on the structures and performances of PVDF/HAPNPs blend membranes. The optimal membrane M2 with 2 wt% HAPNPs exhibited excellent hydrophilicity, outstanding mechanical strength of 19.60 MPa, and high water flux of (2466 +/- 31) Lm(-2)h(-1). The maximum static adsorption capacity of the optimal membrane was about 10.83 mg/g, which is 3.75 times that of the pristine PVDF membrane (2.89 mg/g). PVDF/HAPNPs membranes were not only utilized for static adsorption, but also applied to dynamic dye removal. The possible adsorption mechanism between Congo red (CR) and HAPNPs embedded on the blend membranes was firstly discussed in this work. HAPNPs interacted with CR via Lewis reaction, hydrogen bond interaction, as well as electrostatic attraction to achieve the adsorption effect. Herein, the PVDF/HAPNPs blend membranes with extraordinary hydrophilicity, mechanical strength, and dye removal efficiency possess tremendous potential for practical applications of wastewater treatment.

Abstract :
A novel hydroxyapatite-embedded monolithic column has been facilely prepared in a stainless-steel column with inner diameter of 2.1 mm by the strong adhesion of urea-formaldehyde (UF) resin and exploited as a sorbent for selective on-line solid-phase extraction (on-line SPE) of adenosine triphosphate and its phosphorylated metabolites. The composition for this preparation, including the amount of hydroxyapatite nanopowders and the porogen were investigated to obtain a suitable monolith with large surface area and satisfactory permeability. Owing to anion exchange interaction of hydroxyapatite and hydrophilic interaction of UF monolithic matrix, the prepared monolith showed good extraction efficiency and selectivity towards these phosphorylated analytes. Several parameters for on-line SPE, including ACN percentage in the sampling solution, collection time span, salt concentration of the eluent, sampling and elution flow rate, were optimized with respect to the extraction efficiencies of the target compounds. Under the optimized conditions, the LODs of the analytes were in the range of 0.01-0.04 mu g/g, the recoveries in the spiked samples ranged from 78.3%-92.5% with RSDs < 4.7%. Due to the excellent extraction ability towards phosphorylated compounds in practical samples, a simple on-line SPE-HPLC method using hydroxyapatite-embedded monolith as sorbent has been proposed for monitoring freshness of grass carp.

Based on our developed sodium hyaluronate-functionalized urea-formaldehyde (HA-UF) monolith, a silver nanoparticles-coated monolithic column has been fabricated via the interaction between silver nanoparticles (Ag NPs) and HA. The successful coating of Ag NPs on the parent monolith was proven by SEM, EDAX, UV-vis spectrum and XPS. Nitrogen adsorption desorption isotherms and Barret-JoynerHalenda (BJH) pore size distributions of the parent and resultant monolith were also performed. Due to the Ag+-like affinity interaction caused by the contact of unsaturated compounds and Ag NPs, the Ag NPs-coated monolith showed satisfactory extraction efficiency towards these compounds, and was applied for in-tube solid-phase microextraction (SPME) of monounsaturated fatty acid methyl esters (MUFAMEs). Several factors for in-tube SPME, such as ACN percentage in the sampling solution, elution volume, sampling and elution flow rate, were investigated with respect to the extraction efficiency of model MUFAMEs. Under the optimized SPME conditions, a simple in tube SPME-Ag+-HPLC method for detection of model MUFAMEs has been proposed, the limits of detection (LODs) were less than 5.2 mu g/kg, and the recoveries of spiked French fry samples were ranged from 86.6% to 96.1% with relative standard deviations (RSDs) less than 5.2%. This study provided an Ag NPs-coated monolith with good reproducibility and repeatable extraction performance, and developed an efficient method for in-tube SPME of MUFAMEs in practical food samples. A facile and versatile approach for the preparation of super-hydrophilic, excellent antifouling and hemocompatibility membranes had been developed through the generation in situ of bio-inspired poly-dopamine (PDA) microspheres on PVDF membranes. SEM images showed that the PDA microspheres were uniformly dispersed on the upper surface and the lower surface of the modified membranes. And there were a great number of PDA microspheres immobilized on the cross-section, but the interconnected pores structure was not destroyed. These facts indicated the existence of membrane micro-reactor effect for the whole membrane structure. Considering the remarkable improvement of hydrophilicity, antifouling properties, and permeation fluxes, we also proposed the cluster phenolic hydroxyl effect for the PVDF/PDA hybrid membranes. And the cluster phenolic hydroxyl effect can be ascribed to the all directions distributed phenolic hydroxyl groups on the whole membrane structure. Besides, the self-driven filtration experiments showed the great wetting ability and permeability of the PVDF/PDA hybrid membranes in filtration process without any external pressure. This implied the existence of accelerating self-driven force after the water flow flowed into the internal of membranes, which contributed to the increase of water flow velocity. All the three aspects were in favor of the enhancement of hydrophilicity, antifouling properties and permeability of the modified membranes. Moreover, the conventional filtration tests, oil/water emulsion filtration tests and protein adsorption tests were also carried out to discuss the practical applications of PVDF/PDA hybrid membranes. And the hemocompatibility of the modified membranes was also proved to enhance greatly through the hemolysis tests and platelet adhesion tests, indicating that the membranes were greatly promising in biomedical applications. The strategy of material modification reported here is substrate-independent and can be extended to other substrate materials, and allows the development of novel functional membranes through secondary treatments.
