Abstract :
Peptidoglycan recognition proteins (PGRPs) play an important role in the defense against invading microbes via the recognition of the immunogenic substance peptidoglycan (PGN). Bees possess fewer PGRPs than Drosophila melanogaster and Anopheles gambiae but retain two important immune pathways, the Toll pathway and the Imd pathway, which can be triggered by the recognition of Dap-type PGN by PGRP-LCx with the assistance of PGRP-LCa in Drosophila. There are three isoforms of PGRP-LC including PGRP-LCx, PGRP-LCa and PGRP-LCy in Drosophila. Our previous study showed that a single PGRP-LC exists in bumblebees. In this present study, we prove that the bumblebee Bombus lantschouensis PGRP-LC (Bl-PGRP-LC) can respond to an infection with Gram-negative bacterium Escherichia coli through binding to the Dap-type PGNs directly, and that E. coli infection induces the quick and strong upregulation of PGRP-LC, abaecin and defensin. Moreover, the Bl-PGRP-LC exhibits a very strong affinity for the Dap-type PGN, much stronger than the affinity exhibited by the PGRP-LC from the more eusocial honeybee Apis mellifera (Am-PGRP-LC). In addition, mutagenesis experiments showed that the residue His(390) is the anchor residue for the binding to the Dap-type PGN and forms a hydrogen bond with MurNAc rather than meso-Dap, which interacts with the anchor residue Arg(413) of PGRP-LCx in Drosophila. Therefore, bumblebee PGRP-LC possesses exclusive characteristics for the immune response among insect PGRPs.

Abstract :
Carrot is a very popular vegetable and used for culinary and cosmetic purposes. Carrot seeds can be used for treatment of hangovers and stimulating menstruation. In the present study, the carrot seed protein (CSP) extracted from carrot seed (Daucus carota L.) was hydrolysed by four proteases (papain, trypsin, neutrase, and alcalase). Alcalase hydrolysate exhibited the strongest DPPH radical-scavenging activity (DRSA). The optimum hydrolysis condition for the antioxidant peptide production from CSP was obtained using response surface methodology (RSM). The optimum condition was as follows: hydrolysis time of 3.50 h, substrate concentration of 52.8 g/L, and protease dosage of 419.36 U/g, under which DRSA of 82.46% at 2 mg/mL was obtained. the carrot seed protein hydrolysates (CSPHs) were separated using size exclusion chromatography in order to obtain peptides with stronger antioxidant activity. The hydrolysates were fractionated into four peaks, and fractions F3 and F4 with smaller molecular weight showed stronger antioxidant activity. These findings indicated that the success of RSM in optimizing the hydrolysis conditions and the further work in separation of antioxidant peptides in CSPH is feasible. The CSPH exhibites good antioxidant properties and can be used as useful ingredient in foods.

Abstract :
A limited number of xenobiotic-induced inflammatory episodes occur in the human liver and small intestine under normal physiological conditions, although many xenobiotic agents are metabolized in these organs every day. In this study, we found that rifampicin-activated pregnane X receptor (PXR) plays an important role in the suppression of lipopolysaccharide-activated nuclear factor kappa B (NF-kappa B) activity by increasing the expression of the inhibitor of kappa B alpha (I kappa B alpha) in HepG2 cells. Rifampicin did not increase the expression of I kappa B alpha in PXR knockdown HepG2 cells. Furthermore, we found that the activation of PXR could inhibit the lipopolysaccharide-stimulated nuclear translocation of NF-kappa B p50/p65 using electrophoretic mobility shift assay, immunoprecipitation assays, and microscopy. High levels of I kappa B alpha directly interacted with NF-kappa B and prevented its nuclear translocation, thus inhibiting its binding to consensus DNA sequences in nuclei. Xenobiotic-activated tissue-specific PXR might exert anti-inflammatory actions by antagonizing the xenobiotic-induced transcriptional activity of NF-kappa B in the liver and small intestine. The results also showed that activation of PXR arrested the HepG2 cell cycle in the G(0)/G(1) phase and exhibited cancer prevention potential by inhibiting inflammatory reactions in cells.

Abstract :
采用碱提酸沉法从紫苏籽粕中提取蛋白质,进行蛋白酶筛选,发现紫苏籽蛋白经碱性蛋白酶酶解后有最好的抗氧化活性和水解度(DH).在单因素试验的基础上,采取响应面分析方法,并建立数学模型,确定最佳酶解工艺.试验结果表明:各因素影响紫苏籽粗蛋白酶解液DPPH自由基清除率的主次顺序是:加酶量>酶解时间>pH值,最佳条件是:加酶量3 000 U·g-1,酶解时间5.00 h,pH值9.80,温度50℃,此时DPPH自由基清除率为73.64%,说明紫苏籽蛋白质酶解多肽拥有很高的抗氧化活性.

目的:从TLR4/NF-κ B信号通路的角度探讨芪药鸡金粥对化疗后大鼠肠道紧密连接蛋白的影响,进一步明确芪药鸡金粥调控化疗后肠黏膜损伤的作用机制;初步明确芪药鸡金粥不同剂量与相关指标变化间的量效关系.方法:将40只SPF级Wistar大鼠随机分为模型组、高剂量药膳组、常规剂量药膳组、低剂量药膳组和空白组,每组各8只.实验第1天对模型组及各剂量药膳组行5-FU 150mg/kg一次性腹腔注射,复制化疗所致胃肠黏膜损伤大鼠模型,空白组给予等量生理盐水腹腔注射.实验结束采用ELISA法检测各组大鼠肠道TLR4/NF-κ B信号通路蛋白以及大鼠肠道紧密连接蛋白的表达水平.结果:各组大鼠Claudin-1、ZO-1和JAM-1紧密连接蛋白表达水平的差异有统计学意义(P<0.05);各组大鼠Occludin紧密连接蛋白表达水平差异无统计学意义;各组大鼠IκBα和TLR4通路蛋白变化水平的差异有统计学意义(P<0.05);各组大鼠NF-κ Bp50及NF-κ Bp65通路蛋白表达水平差异无统计学意义.结论:芪药鸡金粥可上调化疗后大鼠胃肠道Claudin-1及JAM-1紧密连接蛋白水平,但对Occludin及ZO-1紧密连接蛋白表达的调控作用尚不明确;其对TLR4/NF-κ B信号通路中的IκBα蛋白具有一定的抑制作用,但对该通路TLR4、NF-κ Bp50及NF-κ Bp65蛋白的调控作用尚不明确;不同剂量芪药鸡金粥与相关指标的量效关系尚不明确,仍需进一步研究探讨.

Abstract :
Ultrasonic pretreatment technology was used to improve the efficiency of the salting process of duck eggs. Ultrasound can be able to reduce the viscosity of egg white as well as increasing the speed of salt penetration. The maturity of salted duck egg was affected by the salt concentration, curing temperature and time. A dynamic model was presented to describe the effects of curing conditions on the maturation of salted eggs. The change of salt content in egg yolk followed a first-order chemical reaction, while the egg white modeled a linear regression equation. It was found that the theoretical estimates obtained from the dynamic model are consistent with the experimental results. The model was proved to reflect dynamic characteristics of salt diffusion and mass transfer in the pickling solution and duck eggs. Through the study of the dynamic model, this experiment revealed the changes of egg's salt content in pickling process under different conditions.

Abstract :
IL-6 (interleukin 6) plays an important role in the development and growth of hepatocellular carcinoma (HCC) via both classic signaling and trans-signaling pathways. Soluble gp130 (sgp130) is known to be a natural inhibitor of the trans-signaling pathway. In the present study, our goal was to investigate whether recombinant sgp130 could suppress the initiation and progression of HCC in mouse models. Our results demonstrate that sgp130 induced an apoptosis of HepG2 cells and inhibited the clonogenicity of HepG2 in vitro. Moreover, the IL-6 trans-signaling pathway is significantly suppressed by sgp130 as reflected by the decrease in the level of STAT3 phosphorylation and other inflammatory factors both in vitro and in vivo. In the DEN-induced HCC mouse model, intravenous injection of sgp130 attenuated hepatic fibrosis at 16 weeks and reduced the initiation and progression of primary HCC at 36 weeks. Furthermore, our results also demonstrate that intravenous administration of sgp130 significantly suppressed the growth and metastasis of xenograft human HCC in NOD/SCID mice.

The pregnane X receptor (PXR), a liver and intestine specific receptor has been reported to be related with the repression of inflammation as well as activation of cytochronnosome P450 3A (CYP3A) expression. We examined the effect of PXR on tetrachloromethane (CCl4)-induced mouse liver inflammation in this work. Ginkgolide A, one main component of Ginkgo biloba extracts (GBE), activated PXR and enhanced PXR expression level, displayed both significant therapeutic effect and preventive effect against CCl4-induced mouse hepatitis. siRNA-mediated decrease of PXR expression significantly reduced the efficacy of Ginkgolide A in treating CCl4-induced inflammation in mice. Flavonoids, another important components of GBE, were shown anti-inflammatory effect in a different way from Ginkgolide A which might be independent on PXR because flavonoids significantly inhibited CYP3A11 activities in mice. The results indicated that anti-inflammatory effect of PXR might be mediated by enhancing transcription level of I kappa B alpha through binding of I kappa B alpha. Inhibition of NF-kappa B activity by NF-kappa B-specific suppressor I kappa B alpha is one of the potential mechanisms of Ginkgolide A against CCl4-induced liver inflammation.

试验采用保加利亚乳杆菌(L.bul-garicus)和嗜热链球菌(S.Thermophilus)混合菌制作酸奶,而后采用低温冷冻干燥制得酸奶粉,并对稳定剂配方进行研究.试验结果表明,酸奶配方为:接种量0.14%,蔗糖添加量7%,发酵时间为6h,发酵温度为42℃.正交试验表明,稳定剂的最佳配方为:CMC-Na 0.10%,卡拉胶0.25%,魔芋胶0.30%.此外,在冷冻前添加冷冻保护剂有助于提高乳酸菌的存活率.

以保加利亚乳杆菌(Lactobacillus bulgaricus)和嗜热链球菌(Streptococcus Thermophilus)混合菌种发酵胡萝卜汁和牛奶的混合物制作酸奶,通过正交实验方法优化胡萝卜风味酸奶的最佳发酵工艺,而后采用低温冷冻干燥制得酸奶粉对其速溶特性进行研究.结果表明,胡萝卜风味酸奶的最佳配方为:接种量0.15%,蔗糖添加量7%,胡萝卜汁添加量15%(均为质量分数),发酵温度为42℃,发酵时间为6h.速溶特性研究表明,在发酵后添加10%的麦芽糊精进行冷冻干燥得到酸奶粉,润湿下沉时间更短,说明其具备良好的速溶特性.此外,在冷冻前添加冷冻保护剂有助于改善胡萝卜风味酸奶粉的冲调性以及提高活菌数.
