Abstract :
Dried leech (Whitmania pigra whitman) has been widely used as a traditional animal-based Chinese medicine. Dried leech extracts have been reported to have various biological activities that are often associated with mammalian glycosaminoglycans. However, their presence and possible structural characteristics within dried leech were previously unknown. In this study, glycosaminoglycans were isolated from dried leech for the first time and their structures were analyzed by the combination of Fourier-transform infrared spectroscopy, liquid chromatography-ion trap/time-of-flight mass spectrometry and polyacrylamide gel electrophoresis. Heparan sulfate and chondroitin sulfate/dermatan sulfate were detected in dried leech with varied disaccharide compositions and possess a heterogeneous structure. Heparan sulfate species possess an equal amount of total 2-O-sulfated, N-sulfated and acetylated disaccharides, while chondroitin sulfate /dermatan sulfate contain high content of 4-O-sulfated disaccharides. Also, the quantitative analysis revealed that the contents of heparan sulfate and chondroitin/dermatan sulfate in dried leech varied significantly, with chondroitin/dermatan sulfate being by far the most abundant. This novel structural information could help clarify the possible involvement of these polysaccharides in the biological activities of the dried leech. Furthermore, leech glycosaminoglycans showed a strong ABTS radical scavenging ability, which suggests the potential of leech polysaccharides for exploitation in the nutraceutical and pharmaceutical industries.

Abstract :
It is well known that the saturated structure in the non-reducing end (NRE) of heparin/heparan sulfate (HS) oligosaccharides is lacking ultraviolet (UV) 232 nm absorbance (normally used for specific detection of C4-C5 unsaturated oligosaccharides), which poses a significant challenge for structure analysis. However, we found that saturated heparin oligosaccharides can be detected at UV 232 nm with sufficient sensitivity in our previous research. In order to further study the UV 232 nm absorbance of the saturated structure, four saturated heparin disaccharides were prepared, and then their UV 232 nm absorptions were analyzed by ion-pair reversed-phase liquid chromatography ion-trap/time-of-flight mass spectrometry (RPIP-LC/MS-IT-TOF) with photodiode array detector. The results show that the saturated heparin disaccharides can be detected with sufficient sensitivity by absorbance at UV 232 nm, which still with 7%-40% of UV 232 nm intensity of heparin unsaturated disaccharides. It seems that the UV 232 nm intensity might be affected by the sulfate groups in the disaccharides. In addition, comparing to the common unsaturated heparin/HS disaccharides as references, the disaccharides with GlcNH(3)(+) residues also showed less sensitivity at UV 232 nm. Finally, we simplified the sequencing method of the N-unsubstituted dp6 oligosaccharides by a simple UV detection method, combined with pH 4.0 HNO2 scission and RPIP-LC/MS-IT-TOF analysis. This strategy offers possibilities for sequencing at N-sulfated residues using alternative pH 1.5 HNO2 scission.

Oviductus ranae (O.ran.) has been widely used as a tonic and a traditional animal-based Chinese medicine. O.ran. extracts have been reported to have numerous biological activities, including activities that are often associated with mammalian glycosaminoglycans such as anti-inflammatory, antiosteoperotic, and anti-asthmatic. Glycosaminoglycans are complex linear polysaccharides ubiquitous in mammals that possess a wide range of biological activities. However, their presence and possible structural characteristics within O.ran. were previously unknown. In this study, glycosaminoglycans were isolated from O.ran. and their disaccharide compositions were analyzed by liquid chromatography-ion trap/time-of-flight mass spectrometry (LC-MS-ITTOF). Heparan sulfate (HS)/heparin (HP), chondroitin sulfate (CS)/dermatan sulfate (DS) and hyaluronic acid (HA) were detected in O.ran. with varied disaccharide compositions. HS species contain highly acetylated disaccharides, and have various structures in their constituent chains. CS/DS chains also possess a heterogeneous structure with different sulfation patterns and densities. This novel structural information could help clarify the possible involvement of these polysaccharides in the biological activities of O.ran..

Abstract :
Heparin (Hep) and heparan sulfate (HS) are complex, sulfated GAGs consist of repeating disaccharide units each composed of a uronic acid, either glucuronate or iduronate [the latter sometimes sulfated at C-2, i.e. IdoA2S], and a derivative of glucosamine [either N-acetylglucosamine, N-sulfated glucosamine, or unsubstituted glucosamine] that can be variably O-sulfated. Hep/HS has important biological functions in developmental processes, angiogenesis, blood coagulation, cell adhesion, and tumour metastasis. These involve interactions with a wide variety of proteins, i.e. enzymes, cytokines, growth factors and extracellular matrix proteins through their specific sequences, patterns or densities of sulfation. Mass spectroscopy is now becoming a much more useful tool for the structural analysis of Hep/HS oligosaccharides because of its high detection sensitivity and molecular specificity. MS/MS of heparin/HS oligosaccharides can produce information-rich glycosidic and cross-ring product ions which can be used to determine the sites of acetylation or sulfation. In particular, LC-MS clearly has the potential now to sequence most oligosaccharides, especially with the recent development of a computational framework for dealing with the complex data generated. In this study, porcine intestines heparin was digested by heparinase I. The obtained crude tetrasaccharides (dp4s) were separated and further purified by strong anion-exchange HPLC and size-exclusion HPLC. The structures of dp4s were then investigated by the combination of disaccharide analysis, reversed-phase liquid chromatography/electrospray ionization ion trap/time-of-flight mass spectrometry (RP-LC/ESI-IT/TOF MS) and a computational simulation method. The results showed that nine tetrasaccharides (dp4s) were obtained, in which two series of dp4s isomer were existed. The dp4 isomers were successfully separated using IPRP-LC/MS system with hexylamine. One series of dp4s contains HexA(2S)-GlcNS and HexA(2S)-GlcNS(6S) disaccharides, and other ones possess HexA-GlcNS(6S) and HexA(2S)-GlcNS(6S). Because MSn analysis could produce abundant structurally useful fragments, the former dp4 isomers were selected to investigate fragmentation patterns for the further potential structure analysis by MS. MS2 analysis indicated that one dp4 was HexA(2S)-GlcNS- HexA(2S)-GlcNS(6S) which had one glycosidic cleavage Y1, two cross-ring cleavages 0,2A2 and 0,3A2 in the first glucosamine residues (from the non-reducing end) of oligosaccharide. In contrast, the other dp4 was HexA(2S)-GlcNS(6S)-HexA(2S)-GlcNS which contained two glycosidic cleavages B2 and Y1, two cross-ring cleavages 1,4A2 and 0,3A2 in the first glucosamine residues, and a cross-ring cleavages 1,5X4 in the first glucuronic acid residues of the oligosaccharide. This study provides a simple and efficient method for identifying the sequences of the Hep/HS oligosaccharide isomers, which might lead to a greater understanding of the biological roles of Hep/HS oligosaccharides in organisms. © 2020, Editorial Board of Journal of Chinese Mass Spectrometry Society. Colla corii asini (CCA) made from donkey-hide has been widely used as a traditional animal-based Chinese medicine. Chondroitin sulfate (CS), dermatan sulfate (DS) and hyaluronic acid (HA) are structurally complex classes of glycosaminoglycans (GAGs) that have been implicated in a wide range of biological activities. However, their possible structural characteristics in CCA are not clear. In this study, GAG fractions containing CS/DS and HA were isolated from CCA and their disaccharide compositions were analyzed by high sensitivity liquid chromatography-ion trap/time-of-flight mass spectrometry (LC-MS-ITTOF). The result showed that CS/DS/HA disaccharides were detected in the three lower salt fractions from anion-exchange chromatography. The sulfation patterns and densities of CS/DS chains in these fractions differed greatly, while HA chains varied in their chain lengths. The quantitative analysis first revealed that the amount of GAGs in CCA varied significantly in total and in each fraction. This novel structural information could help clarify the possible involvement of these polysaccharides in the biological activities of CCA. © 2020, Springer Science+Business Media, LLC, part of Springer Nature.

Alkaline-earth dihalide nanocrystals (NCs) such as SrFCl, owing to their high chemical stability and low phonon energy, are excellent host materials for lanthanide (Ln(3+)) doping to achieve desirable optical properties for various bioapplications. Herein, we report a novel strategy for the synthesis of sub-10 nm Ln(3+)-doped SrFCl NCs with efficient upconverting and downshifting luminescence through a facile one-step hot-injection method. Utilizing the temperature-dependent upconverting luminescence (UCL) from the thermally coupled H-2(11/2) and S-4(3/2) levels of Er3+, we showed the potential of SrFCl:Yb, Er NCs as sensitive UCL nanoprobes for non-contact thermal sensing with a maximum detection sensitivity of 0.0066 K-1, which is among the highest values for thermal sensing based on Er3+-activated UCL nanoprobes. Furthermore, by employing the intense downshifting luminescence from Tb3+ and Eu3+, we demonstrated the successful use of biotinylated SrFCl:Ce, Tb and SrFCl:Eu3+ nanoprobes for biotin receptor-targeted cancer cell imaging, thus revealing the great promise of SrFCl:Ln(3+) nanoprobes for versatile bioapplications. Heparan sulfate (HS)/heparin is a structurally complex class of glycosaminoglycans (GAGs) that have been implicated in a wide range of biological activities. However, their presence within CCA, and their possible structural characteristics, were previously unknown. In this study, GAG fractions containing HS/heparin were isolated from CCA and their disaccharide compositions were analyzed by high sensitivity liquid chromatography-ion trap/time-of-flight mass spectrometry (LC-MS-ITTOF). This revealed that, in addition to the eight commonly seen HS disaccharides, the four rare N-unsubstituted disaccharides were also detected in significant quantities. The disaccharide compositions varied significantly between HS/heparin fractions indicating chains with differing domain structures. This novel structural information may lead to a better understanding of the biological activities (i.e. anticoagulation and antitumor action) of CCA. © 2019, Springer Science+Business Media, LLC, part of Springer Nature.
