Abstract:

通过优化超声波酶辅助法提取绿萝多糖工艺参数,达到提高绿萝多糖得率的目的。以新鲜的天南星科绿萝为原料,干燥、粉碎制备绿萝干粉,将其浸水经超声波及复合酶(木瓜蛋白酶∶果胶酶=1:∶1)处理后,采用3, 5-二硝基水杨酸法测定绿萝多糖得率。研究酶解时间、超声波功率、酶解温度三个单因素对绿萝多糖得率的影响,在此基础上,利用响应面法优化超声波酶辅助法提取绿萝多糖工艺参数。最佳参数为:酶解时间3h,超声波功率76W,酶解温度58℃,测得绿萝多糖得率1.0286%±0.0154%,实际测定值较理论预测值低0.0058%。此方法与传统的热水浸提法相比,大大提高了绿萝多糖得率且用时较短,超声波酶辅助法提取绿萝多糖工艺可为绿萝多糖的开发及应用提供实验基础。
