Abstract:

本发明涉及一种褐藻胶裂解酶突变体及应用,属于酶工程和基因工程领域,该突变体在序列SEQ ID NO.3基础上将第226位E突变为K,经过表达,突变体酶活提高1.11倍,粗酶液经过纯化后比活力较截短酶AlgL‑T157N提高1.03倍;突变体最适温度55℃,在pH 6.0‑8.0内保温1 h酶活力基本不变,具有较强pH稳定性;突变体对褐藻酸钠、Poly M和Poly G的催化效率Kcat/Km分别比AlgL‑T157N提高10.22、8.59、2.97倍。本发明采用结构序列分析和软件辅助筛选相结合的方法,确定突变位点,通过PCR定点突变得到催化活性提高的突变体,为其进一步的工业化应用奠定基础。
