Abstract:

本发明涉及一种人源程序性死亡受体hPD‑1单克隆抗体的制备方法,属于抗体的制备方法技术领域。包括以下步骤:hPD‑1胞外段重组基因的构建;hPD‑1胞外段蛋白的制备;将制备好的抗原注射到Balb/c小鼠体内;再加强免疫3次后采血,ELISA检测血清效价;取效价达到要求的Balb/c小鼠的脾细胞与骨髓瘤细胞进行融合;杂交瘤细胞的克隆化培养;将克隆化培养、筛选获得的杂交瘤细胞接种于Balb/c小鼠腹腔内,即可获得抗体浓度很高的腹水单抗;利用Protein A填料纯化单克隆单体。本发明的方法制备的hPD‑1的单克隆抗体在免疫组化的结果上具有较强的膜定位和具有高特异性、敏感度高、纯度高的优点。
