Abstract:

以卡那霉素提取工艺产生的结晶母液为原料,通过比较不同类型阳离子交换树脂的吸附性能,筛选出卡那霉素B富集率高和杂质吸附率较低的D4阳离子交换树脂.采用多因素多水平的正交试验设计,确定了较佳的吸附富集条件为:上柱结晶母液pH7.0,上柱母液浓度1.26万u/ml,吸附流速1.5V/(v@h);进一步确定了较佳的除杂质洗脱条件为:硫酸铵溶液浓度1.5%(v/v),pH7.5,流速1.75V/(v@h).通过在阳离子交换树脂上的吸附富集和除杂质处理后,再用4%氨水将卡那霉素洗脱下来,将洗脱液浓缩,再经过A1阴离子交换树脂层析分离,可获得高纯度的卡那霉素A、B成品.
