Abstract:
木聚糖酶来源广泛,目前发现的产酶菌株大部分来源于陆地,而对海洋微生物的研究较少.从福建平潭海域筛选到1株产木聚糖酶活力较高的B659菌株,经16SrDNA鉴定为同温层芽胞杆菌.对B659菌株进行产酶培养基及发酵条件的优化.优化的产酶液体培养基是:麸皮6%,豆饼粉1%,K2HPO4 10 mmol/L,初始pH 6.0.经培养基优化的木聚糖酶活力提高了1.6倍.优化的发酵条件是:温度30℃,摇床转速230 r/min,装液量25 mL/250 mL,接种量8%(体积分数).酶活力较优化前提高了0.7倍.经产酶培养基及发酵条件的优化,木聚糖酶活力达到2 838 U/mL,提高了3.4倍,发酵周期由72 h缩短至30 h.
