Translated Abstract
Objective: Studing on the content variation and existential form of Glycyrrhizic acid in Maxing Shigan Decoction by HPLC-Zetasizer Nano.Method: The method was achieved on an Daisogel-C18 column(250 mm×4.6 mm ,5 μm) utilizing methanol 0.2 mol·L-1 solution of ammonium acetate-glacial acetic acid(67∶33∶1) as the mobile phase at the flow rate of 1.0 mL·min-1.The temperature of column was set at 25 ℃.Determining the content variation and existential form of glycyrrhizic acid in the process of Maxing Shigan Decoction in different decocting time, before and after ultrafiltration, rearching the forms of glycyrrhizic acid with Zetasizer Nano.Results: The content of glycyrrhizic acid in Maxing Shigan Decoction significantly rose in the first 20 min, but not apparent changed later.The average particlesize of the decoction was about 250 nm before ultrafiltration, and the glycyrrhizic acid content was 82.4 μg·mL-1.There was no significant particlesize after ultrafiltration, almost no glycyrrhizic acid in the decoction.Conclusion: The content of glycyrrhizic acid in Maxing Shigan Decoction remain unchanged after the increased within the extension time, existing in molecular weight greater than 30 kDa components ,which existed in the form of the compound micelle by supra-molecularinte raction with other ingredients.
